Secretion of neutral and acid DNases in cultivated human lymphocytes after incubation with DNA; possible consequences for inhalation anesthesia.

Abstract After incubation with DNA human lymphocytes release neutral and acid DNase activities into the culture medium; the release depends on DNA concentration and time of cultivation. The electrophoretic mobility of the released neutral DNase activity is in accordance with DNase I and the electrophoretic mobility of the released acid DNase activity with DNase II. The released DNase activities do not originate from dead cells and are not influenced by blast cell formation. The anesthetic halothane can inhibit the released neutral and acid DNase activities. Inhalation anesthesia can possibly disturb the correlation between DNA and DNases in human blood.

Metabolism of propafenone and verapamil by cryopreserved human, rat, mouse and dog hepatocytes: comparison with metabolism in vivo

In the present study we examined the metabolism of [(14)C]propafenone (P) and [(14)C]verapamil (V) using cryopreserved human, dog (Beagle), rat (Sprague-Dawley) and mouse (NMRI) hepatocytes. The percentage ratios of the metabolites were identified after extraction by HPLC with UV and radioactivity detection. Phase-II metabolites were cleaved using beta-glucuronidase. Metabolism of the drugs by cryopreserved hepatocytes was compared with that in the respective species in vivo. Characterization of organic matter in topsoils under rain forest and pasture in the eastern Brazilian Amazon basin

In topsoils under forest and 7-, 12- and 17-year-old pastures, organic matter was characterized by analysing C and N distribution in particle-size fractions, the C decomposition rates of soil and particle-size fractions and by employing density-fractionation of macro-organic matter (>150 μm). The C and N associated with clay fractions increased with increasing age of pasture. The weight (%) of macro-organic matter and its heavy fractions (>1.37 g cm–3) also increased with increasing age of pasture. Production of a Recombinant Catechol 2,3-Dioxygenase for the Degradation of Micropollutants

Phenolic compounds such as catechol represent a particular type of micropollutant whose high stability prevents rapid decay and metabolization in the environment. We successfully cloned a catechol 2,3-dioxygenase (C2,3O) from Pseudomonas putida mt-2 and expressed it in Escherichia coli BER2566. The biomass isolated from shake-flask fermentations was used to partially purify the enzyme. The enzyme proved unstable in clarified liquid fractions (50 mM Tris buffer, pH 7.6) and lost more than 90% of its activity over 7 h at 25 °C. Relative Frequencies and Significance of Faecal Coliforms as Indicators Related to Water Temperature

Summary The faecal coliforms at different sites of a hypereutrophic lake near Valencia (Albufera) were identified and their relative amounts established along an annual cycle. Using lauryl tryptose broth at 35°C, followed by incubation at 44.4°C in 2% brilliant green bile, Escherichia coli and Klebsiella pneumoniae are practically the only coliforms present. A positive correlation was found between the water temperature and the relative amount of these two coliforms: K. pneumoniae predominates at high water temperatures, whereas E. coli shows preponderance during the cold period. Data from: First records of complete annual cycles in water rails Rallus aquaticus show evidence of itinerant breeding and a complex migration system

In water rails Rallus aquaticus, northern and eastern populations are migratory while southern and western populations are sedentary. Few details are known about the annual cycle of this elusive species. We studied movements and breeding in water rails from southernmost Norway where the species occurs year-round. Colour-ringed wintering birds occurred only occasionally at the study site in summer, and vice versa. Geolocator tracks revealed that wintering birds (n = 10) migrated eastwards in spring to breed on both sides of the Baltic Sea, whereas a single breeding bird from the study site wintered in north Italy.
