Isolation of a novel LPS-induced component of the ML superfamily in Ciona intestinalis

ML superfamily represents a group of proteins playing important roles in lipid metabolism and innate immune response. In this study, we report the identification of the first component of the ML superfamily in the invertebrate Ciona intestinalis by means of a subtractive hybridization strategy. Sequence homology and phylogenetic analysis showed that this protein forms a specific clade with vertebrate components of the Niemann-Pick type C2 protein and, for this reason, it has been named Ci-NPC2. This article provides an overview of the immune system in poriferans at a tissue, cellular, and molecular level. Despite their simple organization, sponges have developed molecular mechanisms to recognize and resist foreign organisms. They are able to distinguish among food organisms, pathogens, and sponge-associated organisms. Although they lack specialized immune cells, sponges display molecular precursors, which are similar to molecular mediators involved in innate and adaptive immune systems, present in more evolutionarily advanced taxa, as outlined in succeeding articles in the Phylogeny section.

Staphylococcus aureus alpha-toxin is the archetype of bacterial pore forming toxins and a key virulence factor secreted by the majority of clinical isolates of S. aureus. Toxin monomers bind to target cells and oligomerize to form small beta-barrel pores in the plasma membrane. Many nucleated cells are able to repair a limited number of lesions by unknown, calcium-independent mechanisms. Here we show that cells can internalize alpha-toxin, that uptake is essential for cellular survival, and that pore-complexes are not proteolytically degraded, but returned to the extracellular milieu in the context of exosome-like structures, which we term toxosomes.
