AbstractProcessing of the nonstructural polyprotein of the hepatitis C virus (HCV) requires the serine-type proteinase located in the amino-terminal domain of NS3. To identify residues within NS3 determining substrate specificity, a mutation analysis was performed. Using sequence alignments and three-dimensional structure predictions, amino acids assumed to be important for specificity were replaced and the enzymes were tested in an intracellulartrans-processing assay for their effects on cleavage of an NS4B-5B substrate. Amino acid profiles, established by direct infusion mass spectrometry, have been used to classify vegetable oils according to their botanical origin. The proteins present in hazelnut, sunflower, corn, soybean, olive, avocado, peanut and grapeseed oils were precipitated with acetone, and the residue was hydrolyzed in acid medium, diluted in a hydrochloric acid/ethanol mixture, and infused into the mass spectrometer. The selectivity and sensitivity of the proposed methodologies are achieved by using a specific derivatizing reagent. The different procedures assayed are based on air sampling followed by derivatization of the analytes with OPA and isoleucine in alkaline solution. The derivatization products are separated and determined by liquid chromatography and fluorescence detection. In questi ultimi anni, il commercio di cappero, a livello internazionale, ha fatto registrare un trend positivo rappresentato da uno scambio di prodotto pari, secondo dati forniti dal COMTRADE, a circa 3 mila tonnellate. A fronte di tali incrementi, in Italia, la superficie investita a cappero è scesa drasticamente, passando da circa 1000 ha nel ’83 a circa 170 ha nel 2006. Ciò, presumibilmente, è dovuto alla carente attuazione di sapienti strategie di marketing sul prodotto cappero da parte degli imprenditori agricoli. The 17O NMR chemical shifts of anisole and all 19 chlorinated anisoles measured in CDCI3 at ambient temperature show a dispersion of approximately 40 ppm. The substituent chemical shifts (SCS) of di- or more highly chlorinated compounds cannot be reproduced by addition of single substituent effects. Theoretical calculations of the 17O NMR chemical shifts have been attempted by molecular mechanical and semi-empirical molecular orbital methods at the INDO level, but the results are not satisfactory.
