Mitochondrial and chromosomal DNA alterations in human chromophobe renal cell carcinomas

1992

Renal cell tumours are characterized by the loss of chromosome 3p and trisomy of 5q segments (common, non-papillary renal cell carcinoma), or by trisomy of chromosomes 7 and 17 and loss of the Y chromosome (papillary renal cell carcinoma), or by random karyotype changes and mitochondrial DNA alterations (renal oncocytoma). We have studied by means of RFLP analysis the genomic and mitochondrial DNA in 11 chromophobe renal cell carcinomas, which have a unique morphology among kidney cancers. We found a loss of the constitutional heterozygosity at chromosomal regions 3p, 5q, 17p, and 17q, a combination of allelic losses that has not been found in other types of renal cell tumours. Three of the…

Caki-1 Cells Represent an in vitro Model System for Studying the Human Proximal Tubule Epithelium

2007

Background/Aims: The human proximal tubule (PT) epithelium is distinguished from other nephron segments via several unique characteristics. Studies assessing PT epithelium increasingly employ cell lines, bypassing the complexity of primary cell cultures. However, few human model systems exist for studying PT cells in vitro. The current work involves an intensive characterization of Caki-1 cells, a commercially available human renal cell line. Methods: Caki-1 cells were validated as a representative model system for PT cell research via morphological, physiological and biochemical investigations including light and transmission electron microscopy, trans…

Interrelationship between mean arterial blood pressure, blood flow, and vascular resistance in solid tumor tissue of DS-carcinosarcoma

1975

Bone Metastasis in Renal Cell Carcinoma is Preprogrammed in the Primary Tumor and Caused by AKT and Integrin α5 Signaling

2015

Bone metastasis develops in 30% of all patients with renal cell carcinoma. We elucidated the mechanisms that lead to and predict bone metastasis of renal cell carcinoma.Nine renal cell carcinoma primary cell lines and 30 renal cell carcinoma tissue specimens (normal and tumor tissue) were collected from 3 patients with no metastasis and 10 with lung or bone metastasis within 5 years after nephrectomy. Cell migration was analyzed in a Boyden chamber and proliferation was assessed by bromodeoxyuridine incorporation. Adhesion to fibronectin, and collagen I and IV was determined after cell staining. The expression and/or activity of cellular signaling molecules was quantified by Western blot.Co…

Vaginal Metastasis from Renal Cell Carcinoma: The Role of Electron Microscopy

1989

A case of vaginal metastasis from a renal cell carcinoma is reported. This rare localization is often the first clue leading to the diagnosis of an occult renal adenocarcinoma. The role of electron microscopy is emphasized, since the histologic aspect of the lesion is similar to that of a primary vaginal mesonephric adenocarcinoma.

Wilms tumours diagnosed by routine physical examinations.

1988

Sir: The prognosis of stage I and -II Wilms tumours with favourable histology is good, reaching 97% (stage I) and 92% (stage II) 5 year disease-free survival rates [1]. With these figures in mind the question arises whether the overall prognosis of Wilms tumours could be improved by presymptomatic routine physical examinations in an attempt to avoid stage III to V primary diagnoses. In the Federal Republic of Germany six routine physical examinations (RPE) are offered to infants, and two additional ones to children between 12 and 48 months. The 1980-1987 German Wilms tumour study [1] listed 313 patients, 29 of whom (9%) were diagnosed by such routine examinations. As shown in Table 1, the R…

Hemodilution in isolated tumor perfusion.

1984

Experiments are performed to study the influence of local hemodilution on tumor blood flow, oxygen availability in tumor tissue and O2 consumption of cancer cells. The results obtained clearly show that hemodilution in isolated tumor perfusion can distinctly improve nutritive blood flow through solid tumors. This can be utilized to enhance pharmacokinetics of antitumor drugs. Due to the improved metabolic status, the pharmacodynamics of some antitumor drugs should also be enhanced. To achieve a maximum improvement of the O2 supply to the tumor, hematocrit values should not be decreased below 0.20.

Identification of metabolic enzymes in renal cell carcinoma utilizing PROTEOMEX analyses

2003

Abstract PROTEOMEX, an approach which combines conventional proteome analysis with serological screening, is a powerful tool to separate proteins and identify immunogenic components in malignant diseases. By applying this approach, we characterized nine metabolic enzymes which were differentially expressed in renal cell carcinoma (RCC) cell lines and compared their expression profiles to that of normal kidney epithelium cells. Four of these proteins, superoxide dismutase (SODC), triosephosphatase isomerase (TPIS), thioredoxin (THIO) and ubiquitin carboxyl-terminal hydrolase (UBL1) were further analysed for both their constitutive and interferon (IFN)-γ inducible protein expression pattern i…

B7/CD28 costimulation of T cells induces a distinct proteome pattern.

2005

Effective immune strategies for the eradication of human tumors require a detailed understanding of the interaction of tumor cells with the immune system, which might lead to an optimization of T cell responses. To understand the impact of B7-mediated costimulation on T cell activation comprehensive proteome analysis of B7-primed T cell populations were performed. Using this approach we identified different classes of proteins in T cells whose expression is either elevated or reduced upon B7-1- or B7-2-mediated CD28 costimulation. The altered proteins include regulators of the cell cycle and cell proliferation, signal transducers, components of the antigen processing machinery, transporters…

Detection of renal cell carcinoma-associated markers via proteome- and other 'ome'-based analyses

2004

Proteome analysis has rapidly developed in the post-genome era and is now widely accepted as the complementary technology for genetic profiling. It has been shown to be a powerful tool for studying human diseases and for identifying novel prognostic, diagnostic and therapeutic markers. This review focuses on the identification of new biomarkers and therapeutic targets for renal cell carcinoma using different 'ome'-based technologies.
