Novel molluskan biomineralization proteins retrieved from proteomics: a case study with upsalin.

12 pages; International audience; The formation of the molluskan shell is regulated by an array of extracellular proteins secreted by the calcifying epithelial cells of the mantle. These proteins remain occluded within the recently formed biominerals. To date, many shell proteins have been retrieved, but only a few of them, such as nacreins, have clearly identified functions. In this particular case, by combining molecular biology and biochemical approaches, we performed the molecular characterization of a novel protein that we named Upsalin, associated with the nacreous shell of the freshwater mussel Unio pictorum. Analysis of enniatins and beauvericin by LC-MS/MS in wheat-based products

Due to the matrix complexity for wheat-based products, a comparative study of different rapid extraction procedures was performed for the extraction of enniatins (ENA, ENA1, ENB, ENB1) and beauvericin in flour, pasta, breakfast cereals, and biscuits. Three different approaches were studied during the extraction and purification steps (shaker, Ultra-Turrax, and QuEChERS) for each matrix. Liquid chromatography coupled to tandem mass spectrometry (LC-MS/MS) with electrospray source working in a positive mode was used. Representative major element compositions of clinopyroxenes and phlogopites from ultrapotassic rocks from Nangqian basin (Eastern Tibet)

Representative major element compositions (in wt%) of clinopyroxenes and phlogopites from Nangqian ultrapotassic rocks. In-situ major-element compositions of mineral phases were obtained using the JEOL JXA-8230 Electron Microprobe at ISTerre, University Grenoble Alpes. Analytical conditions were 15 kV accelerating voltage and 12 nA beam current. The ZAF procedure was applied to reduce the raw data. The microprobe was calibrated using natural and synthetic standards. An X-ray element map of a calcite-bearing aggregate was acquired using 15 kV accelerating voltage and 10 nA beam current.

Sea urchin embryos as a model system for studying autophagy induced by cadmium stress

It is well known that sea urchin embryos are able to activate different defense strategies against stress. We previously demonstrated that cadmium treatment triggers the accumulation of metal in embryonic cells and the activation of defense systems depending on concentration and exposure time, through the synthesis of heat shock proteins and/or the initiation of apoptosis. Cadmium induces an apoptotic response in sea urchin embryos.

Cadmium is a heavy metal toxic for living organisms even at low concentrations. It does not have any biological role, and since it is a permanent metal ion, it is accumulated by many organisms. In the present paper we have studied the apoptotic effects of continuous exposure to subacute/sublethal cadmium concentrations on a model system: Paracentrotus lividus embryos. We demonstrated, by atomic absorption spectrometry, that the intracellular amount of metal increased during exposure time. We found, using terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling assay, that long treatments with cadmium triggered a severe DNA fragmentation. A novel wingless gene was isolated from the marine colonial hydroid Hydractinia echinata. Alignments and Bayesian inference analysis clearly assign the gene to the Wnt5A group. In line with data found for the brachyury ortholog of Hydractinia, He-wnt5A is expressed during metamorphosis in the posterior tip of the spindle-shaped planula larva, suggesting that the tip functions as a putative organizer during metamorphosis. Additionally, the outermost cells of the posterior tip are omitted from apoptosis during metamorphosis. We introduce a novel research proposal project aimed to build a robotic setup in which the Telenoid learns to improvise jazz singing in a duet with a human singer. In the proposed application, the Telenoid acts in teleoperated mode during the learning phase, while it becomes more and more autonomous during the working phase.
