Improving appropriateness in laboratory medicine: How, when and why

2019

The issue of the appropriateness in laboratory medicine has been discussed from several years in association to the parallel onset of two aspects: 1) the significant increase in tests demand and utilization, thanks to the development of laboratory automation and information laboratory systems (LIS), that allow to provide timely and reliable results to clinicians; 2) the opportunity, thanks to new pathophysiological knowledge and new technologies to introduce new and more sophisticated tests in clinical practice, providing a relevant support to the clinician in the management of patients, according to the improved vision of personalized medicine. As a consequence, the potential inappropriate…

Radioaktiivisten päästöjen mittaaminen ilmanvaihdon poistoilmasta

2016

Mittaus toteutettiin MAP Medical Technologies Oy:n (MAP) vanhalla päästömittausjärjestelmällä tuottamalla siihen uusi laskentaohjelmisto. Tuloksia vertailtiin MAP:n uuden järjestelmän tuottamiin tuloksiin sekä erikoistyössä laskettuihin päästörajoihin. Vertailussa todettiin, että eri järjestelmillä…

The C-X-C Motif Chemokine Ligand 1 Sustains Breast Cancer Stem Cell Self-Renewal and Promotes Tumor Progression and Immune Escape Programs

2021

Breast cancer (BC) mortality is mainly due to metastatic disease, which is primarily driven by cancer stem cells (CSC). The chemokine C-X-C motif ligand-1 (CXCL1) is involved in BC metastasis, but the question of whether it regulates breast cancer stem cell (BCSC) behavior is yet to be explored. Here, we demonstrate that BCSCs express CXCR2 and produce CXCL1, which stimulates their proliferation and self-renewal, and that CXCL1 blockade inhibits both BCSC proliferation and mammosphere formation efficiency. CXCL1 amplifies its own production and remarkably induces both tumor-promoting and immunosuppressive factors, includingSPP1/OPN,ACKR3/CXCR7,TLR4,TNFSF10/TRAILandCCL18and, to a lesser exte…

A perspective analysis: microRNAs, glucose metabolism, and drug resistance in colon cancer stem cells

2021

Metabolism sustains the stemness of Cancer Stem Cells (CSCs), affecting, in turn, tumor heterogeneity, metastatic potential, and therapy resistance. Therefore, it is appealing to target CSCs metabolism as a new therapeutic approach. Consequently, we paid considerable attention to the anti-apoptotic microRNA miR-483-3p, that we reported being regulated by glucose metabolism in liver cancer cells. We investigated the therapeutic potential of targeting miR-483-3p by using the anti-glucose metabolism 2-deoxyglucose (2-DG) molecule in tumor Xenograft mouse model originating from two different Colon-Cancer Stem Cell lines (CCSC lines). We show that 2-DG treatment does not affect CCSCs during tumo…

Adipose stromal cells promote the transition of colorectal cancer cells toward a mesenchymal-like phenotype

2021

ABSTRACT Colon cancer progression is among the risks that increase with obesity. We have recently unveiled the molecular mechanism by which adipose tissue-released molecules, HGF and IL-6, make colorectal cancer (CRC) cells acquiring mesenchymal traits. Targeting of adipose-derived factors abrogate the metastatic potential of CRC stem cells (CR-CSCs) in obese patients.

Effective targeting of breast cancer stem cells by combined inhibition of Sam68 and Rad51

2022

AbstractBreast cancer (BC) is the second cause of cancer-related deceases in the worldwide female population. Despite the successful treatment advances, 25% of BC develops resistance to current therapeutic regimens, thereby remaining a major hurdle for patient management. Current therapies, targeting the molecular events underpinning the adaptive resistance, still require effort to improve BC treatment. Using BC sphere cells (BCSphCs) as a model, here we showed that BC stem-like cells express high levels of Myc, which requires the presence of the multifunctional DNA/RNA binding protein Sam68 for the DNA-damage repair. Analysis of a cohort of BC patients displayed that Sam68 is an independen…

Messing Up the Cancer Stem Cell Chemoresistance Mechanisms Supported by Tumor Microenvironment

2021

Despite the recent advances in cancer patient management and in the development of targeted therapies, systemic chemotherapy is currently used as a first-line treatment for many cancer types. After an initial partial response, patients become refractory to standard therapy fostering rapid tumor progression. Compelling evidence highlights that the resistance to chemotherapeutic regimens is a peculiarity of a subpopulation of cancer cells within tumor mass, known as cancer stem cells (CSCs). This cellular compartment is endowed with tumor-initiating and metastasis formation capabilities. CSC chemoresistance is sustained by a plethora of grow factors and cytokines released by neighboring tumor…
