Caractérisation cytomique de cellules transformées de tabacs exprimant le domaine « hub » de la chaîne lourde de la clathrine

Abstract. Sediments containing gas hydrate dispersed in the pore space are known to show a characteristic seismic anomaly which is a high attenuation along with increasing seismic velocities. Currently, this observation cannot be fully explained albeit squirt-flow type mechanisms on the microscale have been speculated to be the cause. Recent major findings from in situ experiments, using the gas in excess and water in excess formation method, and coupled with high-resolution synchrotron-based X-ray micro-tomography, have revealed the systematic presence of thin water films between the quartz grains and the encrusting hydrate. The data obtained from these experiments underwent an image proce…
Natural gas hydrate investigations by synchrotron radiation X-ray cryo-tomographic microscopy (SRXCTM)
2008
[1] We report the 3D microstructure analyses of natural gas hydrates sampled from Gulf of Mexico. The samples were characterized by synchrotron radiation X-ray cryo-tomographic microscopy (SRXCTM) using the ‘TOMCAT’ beam line at the Swiss Light Source (SLS). The SRXCTM demonstrates its applicability to unlock some microscopic features of the marine hydrates, in particular of crystallite size and grain boundary network. The gas hydrate domains are surrounded by a network of pores of typically a few micrometers, which are largely due to decomposition. Out of the SRXCTM data, the porosity, total volume of the voids, the void surface area and number of the total gas-filled voids have been calcu…
Using growth and geochemical composition of Clathromorphum compactum to track multiscale North Atlantic hydro-climate variability
2020
International audience; Records of ocean/atmosphere dynamics over the past centuries are essential to understand processes driving climate variability. This is particularly true for the Northwest Atlantic which is a key region with an essential role in global climate regulation. Over the past two decades, coralline red algae have been increasingly used as environmental and climatic archives for the marine realm and hold the potential to extend long-term instrumental measurements. Here, we investigate the possibility to extract climate and environmental information from annual growth patterns and geochemical composition of the coralline red algae, Clathromorphum compactum, from Saint-Pierre …
Sorting Motifs Involved in the Trafficking and Localization of the PIN1 Auxin Efflux Carrier
2016
In contrast with the wealth of recent reports about the function of μ-adaptins and clathrin adaptor protein (AP) complexes, there is very little information about the motifs that determine the sorting of membrane proteins within clathrin-coated vesicles in plants. Here, we investigated putative sorting signals in the large cytosolic loop of the Arabidopsis (Arabidopsis thaliana) PIN-FORMED1 (PIN1) auxin transporter, which are involved in binding μ-adaptins and thus in PIN1 trafficking and localization. We found that Phe-165 and Tyr-280, Tyr-328, and Tyr-394 are involved in the binding of different μ-adaptins in vitro. However, only Phe-165, which binds μA(μ2)- and μD(μ3)-adaptin, was found …
Constitutive expression of clathrin hub hinders elicitor-induced clathrin-mediated endocytosis and defense gene expression in plant cells.
2012
International audience; Endocytosis has been recently implicated in the signaling network associated with the recognition of microbes by plants. In a previous study, we showed that the elicitor cryptogein was able to induce clathrin-mediated endocytosis (CME) in tobacco suspension cells. Herein, we investigate further the induced CME by means of a GFP-tagged clathrin light chain and a CME inhibitor, the hub domain of clathrin heavy chain. Hub constitutive expression does affect neither cell growth nor constitutive endocytosis but abolishes cryptogein-induced CME. Such an inhibition has no impact on early events in the cryptogein signaling pathway but reduces the expression of defense-associ…
ABP1 Mediates Auxin Inhibition of Clathrin-Dependent Endocytosis in Arabidopsis
2010
SummarySpatial distribution of the plant hormone auxin regulates multiple aspects of plant development. These self-regulating auxin gradients are established by the action of PIN auxin transporters, whose activity is regulated by their constitutive cycling between the plasma membrane and endosomes. Here, we show that auxin signaling by the auxin receptor AUXIN-BINDING PROTEIN 1 (ABP1) inhibits the clathrin-mediated internalization of PIN proteins. ABP1 acts as a positive factor in clathrin recruitment to the plasma membrane, thereby promoting endocytosis. Auxin binding to ABP1 interferes with this action and leads to the inhibition of clathrin-mediated endocytosis. Our study demonstrates th…
Systematic analysis of specific and nonspecific auxin effects on endocytosis and trafficking.
2021
Abstract The phytohormone auxin and its directional transport through tissues are intensively studied. However, a mechanistic understanding of auxin-mediated feedback on endocytosis and polar distribution of PIN auxin transporters remains limited due to contradictory observations and interpretations. Here, we used state-of-the-art methods to reexamine the auxin effects on PIN endocytic trafficking. We used high auxin concentrations or longer treatments versus lower concentrations and shorter treatments of natural indole-3-acetic acid (IAA) and synthetic naphthalene acetic acid (NAA) auxins to distinguish between specific and nonspecific effects. Longer treatments of both auxins interfere wi…
The Plant Defense Elicitor Cryptogein Stimulates Clathrin-Mediated Endocytosis Correlated with Reactive Oxygen Species Production in Bright Yellow-2 …
2008
Abstract The plant defense elicitor cryptogein triggers well-known biochemical events of early signal transduction at the plasma membrane of tobacco (Nicotiana tabacum) cells, but microscopic observations of cell responses related to these early events were lacking. We determined that internalization of the lipophilic dye FM4-64, which is a marker of endocytosis, is stimulated a few minutes after addition of cryptogein to tobacco Bright Yellow-2 (BY-2) cells. This stimulation is specific to the signal transduction pathway elicited by cryptogein because a lipid transfer protein, which binds to the same receptor as cryptogein but without triggering signaling, does not increase endocytosis. To…
Valuable natural products from marine and freshwater macroalgae obtained from supercritical fluid extracts
2017
The biologically active compounds (fatty acids, pigments, phenolics, and flavonoid content) were studied in supercritical fluid extracts from the biomass of marine (Ulva clathrata, Cladophora glomerata, Polysiphonia fucoides, and their multi-species mixture) and freshwater (C. glomerata) macroalgae. Different extraction techniques were used in order to compare differences in the biologically active compound composition of the macroalgal extracts. The results indicated that the saturated and unsaturated fatty acids ranged from C9:0 to C22:0. The analysis of differences in the composition of unsaturated to saturated fatty acids in extracts showed that palmitic acid (C16:0) and oleic acid (C18…
