Congenital reduction in number of nephrons accelerates renal damage, and limits proximal tubular growth response to release of neonatal partial urete…
2012
Aim: to analyze the evolution of the response to persistent partial ureteral obstruction (PUO) and obstruction/release (PUO/Rel) in mice with 50% reduction in the number of nephrons from 21 days of age (time of weaning) to 42 days of age (adulthood). Methods: C57/BL6 wild-type (WT) and mice with oligosyndactylism and 50% reduction of nephron number (Os/+), were subjected to sham operation or PUO in the first 2 days of life. Additional mice underwent release of obstruction at 7 days (PUO-Rel). The study was performed in kidneys harvested at 21 and 42 days. Using histomorphometry, glomerular area (GA) was determined by PAS staining, and fractional proximal tubular (PT) mass and intact glomeru…

L’impatto della pandemia sui rapporti contrattuali: problemi e rimedi
2021
This paper aims to analyze the global disaster caused by the COVID-19 pandemic that has disrupted the existence of almost all of humanity. In addition, examine the significant limitations limitations on individual and collective freedoms to protect public health and, secondly, to mitigate as much as possible the impact of the pandemic on economic activities. The attention of the article focuses exclusively on the rules that impact on the general discipline of obligations and contracts. The essay deals with the institutions of contract law that are best suited to counteract the effects of the pandemic on contractual relationships, especially those long term. Particular attention is paid to t…
Ethyl Hexanoate Transfer Modeling in Carrageenan Matrices for Determination of Diffusion and Partition Properties.
2007
; Aroma compound properties in food matrices, such as volatility and diffusivity, have to be determined to understand the effect of composition and structure on aroma release and perception. This work illustrates the use of mass transfer modeling to identify diffusion and partition properties of ethyl hexanoate in water and in carrageenan matrices with various degrees of structure. The comparison of results obtained with a diffusive model to those obtained with a convective model highlights the importance of considering the appropriate transfer mechanism. Modeling of the preliminary experimental steps ensures correct estimation of the conditions for the main aroma release step. The obtaine…
Rationale and evidence on the use of tocilizumab in COVID-19: a systematic review
2020
Abstract Background Tocilizumab is an IL-6 receptor-blocking agent proposed for the treatment of severe COVID-19. The aim of this systematic review was to describe the rationale for the use of tocilizumab for the treatment of COVID-19 and to summarize the available evidence regarding its efficacy and safety. Methods MEDLINE, PubMed, EMBASE, pre-print repositories (bioRxiv and medRxiv) and two trial Registries were searched for studies on the use of tocilizumab in COVID-19 or SARS-CoV-2 infection, viral pneumonia, and/or sepsis until 20th June 2020. Results We identified 3 indirect pre-clinical studies and 28 clinical studies including 5776 patients with COVID-19 (13 with a comparison group,…
CMB spectral distortions in generic two-field models
2017
We investigate the CMB $\mu$ distortion in models where two uncorrelated sources contribute to primordial perturbations. We parameterise each source by an amplitude, tilt, running and running of the running. We perform a detailed analysis of the distribution signal as function of the model parameters, highlighting the differences compared to single-source models. As a specific example, we also investigate the mixed inflaton-curvaton scenario. We find that the $\mu$ distortion could efficiently break degeneracies of curvaton parameters especially when combined with future sensitivity of probing the tensor-to-scalar ratio $r$. For example, assuming bounds $\mu < 0.5 \times 10^{-8}$ and $r<0.0…
Benzo[a]pyrene represses DNA repair through altered E2F1/E2F4 function marking an early event in DNA damage-induced cellular senescence
2020
AbstractTranscriptional regulation of DNA repair is of outmost importance for the restoration of DNA integrity upon genotoxic stress. Here we report that the potent environmental carcinogen benzo[a]pyrene (B[a]P) activates a cellular DNA damage response resulting in transcriptional repression of mismatch repair (MMR) genes (MSH2, MSH6, EXO1) and of RAD51, the central homologous recombination repair (HR) component, ultimately leading to downregulation of MMR and HR. B[a]P-induced gene repression is caused by abrogated E2F1 signalling. This occurs through proteasomal degradation of E2F1 in G2-arrested cells and downregulation of E2F1 mRNA expression in G1-arrested cells. Repression of E2F1-me…
Randomized placebo-controlled trial comparing fluticasone aqueous nasal spray in mono-therapy, fluticasone plus cetirizine, fluticasone plus monteluk…
2004
BACKGROUND: Corticosteroids are considered to be particularly effective in reducing nasal congestion and are therefore recommended as first-line treatment in allergic rhinitis patients with moderate to severe and/or persistent symptoms. OBJECTIVE: We compared the clinical efficacy of fluticasone propionate aqueous nasal spray (FPANS) 200 microg given once daily, administered in mono-therapy or combined therapy with a H1 receptor antagonist (cetirizine, CTZ) or with a leukotriene antagonist (montelukast, MSK), and the combined therapy of CTZ plus MSK in the treatment of patients affected by allergic rhinitis to Parietaria during natural pollen exposure. In addition, we examined the effect of…
Nuclear Translocation of Mismatch Repair Proteins MSH2 and MSH6 as a Response of Cells to Alkylating Agents
2000
Mammalian mismatch repair has been implicated in mismatch correction, the prevention of mutagenesis and cancer, and the induction of genotoxicity and apoptosis. Here, we show that treatment of cells specifically with agents inducing O(6)-methylguanine in DNA, such as N-methyl-N'-nitro-N-nitrosoguanidine and N-methyl-N-nitrosourea, elevates the level of MSH2 and MSH6 and increases GT mismatch binding activity in the nucleus. This inducible response occurs immediately after alkylation, is long-lasting and dose-dependent, and results from translocation of the preformed MutSalpha complex (composed of MSH2 and MSH6) from the cytoplasm into the nucleus. It is not caused by an increase in MSH2 gen…
Gene expression is circular: factors for mRNA degradation also foster mRNA synthesis.
2013
SummaryMaintaining proper mRNA levels is a key aspect in the regulation of gene expression. The balance between mRNA synthesis and decay determines these levels. We demonstrate that most yeast mRNAs are degraded by the cytoplasmic 5′-to-3′ pathway (the “decaysome”), as proposed previously. Unexpectedly, the level of these mRNAs is highly robust to perturbations in this major pathway because defects in various decaysome components lead to transcription downregulation. Moreover, these components shuttle between the cytoplasm and the nucleus, in a manner dependent on proper mRNA degradation. In the nucleus, they associate with chromatin—preferentially ∼30 bp upstream of transcription start-sit…
