Systemic lupus erythematosus (SLE) is an autoimmune disease characterized by the emergence of autoreactive T cells. Humans and mice with SLE have reduced numbers of CD1d-restricted invariant natural killer T (iNKT) cells, suggesting a key role for these cells in its immunopathogenesis. This subset uses an invariant TCR constituted by Valpha14 Jalpha281 chains paired with some Vbeta domains. Urodele amphibians have weak and slow immune responses compared to mammals and anuran amphibians. Using new culture conditions, we tested the ability of lymphocytes of a well-studied salamander, the Mexican axolotl (Ambystoma mexicanum) to proliferate in vitro with diverse mitogenic agents. We demonstrated that the axolotl has a population of B lymphocytes that proliferate specifically and with a high stimulation index to the lipopolysaccharide (LPS) known as a B-cell mitogen in mammals. NZB mice develop an autoimmune disease of unknown etiology. Since the detection of immunoregulatory T-cells it has been speculated that disbalances of these cells may be important in the course of the NZB disease. By utilization of monoclonal antibody defining immunoregulatory Lyt subsets and a FACS IV system we investigated whether differences in the number and/or marker densities of given subsets exist between NZB and the normal reference strain BALB/c. Newborn animals and animals up to 60 weeks of age were tested. Only few studies are available on sleep disorders in Angelman syndrome (AS), a neurodevelopmental disorder with several behavior disturbances. The aim of this study was to determine the prevalence of sleep disorders in a relatively large group of AS subjects, compared to that of age-matched controls. The outfit of husbandry facilities of, and the enrichment provided for, experimental rodents plays an important role in the animals’ welfare, and hence also for the societal acceptance of animal experiments. Whether rats and mice benefit from being provided with running wheels or plates is discussed controversially. Here we present observations from a feeding experiment, where rats were provided a running plate. As a pilot study, six identical cages, with three animals per cage, were filmed for six days, and the resulting footage was screened for the number of bouts and the time the animals spent on the plates. Abstract We have previously shown that injection of donor-derived Fischer rat OX62+ dendritic cells plus an anti-CD4 monoclonal antibody generates donor-specific CD4+CD25+FoxP3+ regulatory T cells in Lewis rats spleens. The regulatory T cells indefinitely prolonged the survival of skin graft from Fischer rat and abrogated the antidonor antibody response. We have now shown that an injection of 2 × 10 6 donor-derived OX62+ dendritic cells plus 2 mg nondepleting anti-CD4 monoclonal antibody (W3/25) at 28 days before grafting induced indefinite skin graft survival in this combination; whereas an injection on day −1 prolonged it only to 50 days.
