The Role of Tumor Volume in ‘Reoxygenation’ upon Cyclophosphamide Treatment

The effect of cyclophosphamide (CP) injection (60 mg/kg i.p., single dose) on volume growth and tissue oxygenation (pO2 distribution) was investigated in rat DS-sarcomas. CP was administered 4 days after subcutaneous (s.c.) tumor implantation (volume approximately 0.35 ml). Polarographic pO2 measurements were performed in the subcutis at the hind foot dorsum and in tumors 72 h after CP administration. The oxygenation status of these tissues was compared with that of saline-treated controls. CP-injection caused a mean growth delay of 11 days in DS-sarcomas and had no impact on the oxygenation status of the subcutis. Abstract Using high-performance-liquid-chromatography (HPLC) measurement of acetylcholine, choline acetyltransferase (ChAT) enzyme assay and anti-ChAT immunohistochemistry, we have investigated the expression of the cholinergic system in pineal glands of male rats. Glands procured during the day period (1200 h) contained significant amounts of acetylcholine (0.5 pmol/gland). A similar content was found in pineal glands after a 48 h culture period, i.e. when the intrapineal nerve fibres have degenerated. Genetically obese Zucker (fa/fa) rats were used as an experimental model to study the effects of hypolipidemic agents on peroxisome proliferation; comparison was made with Zucker lean phenotype (Fa/-) and Sprague-Dawley strain/phenotype. The pharmacokinetics of a single administration of ciprofibrate (1 or 3 mg/kg), appeared to be similar in all strains/phenotypes. After a 2-week oral administration at the same dosages, there were dosage-related increases in hepatocellular peroxisomal yield and in the hepatic enzymes' cyanide-insensitive acyl-CoA oxidase and catalase. Hepatocytes entrapped in collagen gel and cultured in serum-free conditions survived longer than cells cultured on plastic (5 days vs. 3 weeks), showed fewer signs of early cell senescence (no increase in c-fos oncoprotein expression), and maintained the expression of differentiated hepatic metabolic functions over a longer period of time. Cells cultured in collagen gels retained their ability to respond to hormones. The insulin-stimulated glycogen synthesis rate remained fairly constant during 18 days in culture (between 5.4 +/- 0.37 and 9 +/- 2.7 nmol glucose/h/microg DNA). Abstract Endplate preparations of the left rat hemidiaphragm were incubated with [ 3 H]choline to label neuronal acetylcholine stores. Elevation of the concentration (13.5–135 mmol/l) of extracellular potassium chloride (KC1) stimulated the release of [ 3 H]acetylcholine in a concentration-dependent manner. KC1 (27 mmol/l) still caused a significant efflux of [ 3 H]acetylcholine in a Ca 2+ -free medium. Antisecretory drugs are effective antiulcer agents, but its chronic use generates hypergastrinemia and accelerates the development of atrophic gastritis in Helicobacter pylori-positive patients. We have recently shown that gastrin exerts a proinflammatory effect in rats through CCK-2 receptor activation that contributes to the inflammation induced by H. pylori. Ontogeny of bradykinin B 2 receptors in the rat kidney: Implications for segmental nephron maturation. Kinins modulate renal function, yet their role in the developing kidney is largely unknown. To explore the developmental role of the kallikrein-kinin system, we examined the postnatal ontogeny and intrarenal localization of B 2 receptors in the rat. Northern blot analysis and RT-PCR documented the expression of B 2 receptor mRNA in the kidney and extrarenal tissues of fetal, neonatal and adult animals. The abundance of B 2 receptor mRNA is 10- to 30-fold higher in neonatal than adult tissues in the following order: kidney > heart > aorta > lung > brain. Receptor autoradiography revealed a …
