Lipopolysaccharides and glucagon-like peptide 1 : from molecular mechanisms to pathophysiology

Obesity and type 2 diabetes are metabolic diseases which have reached epidemic proportions worldwide. These metabolic disorders are related to a low grade inflammation whose molecular origin is still unknown. Previous studies have highlighted the involvement of the gut microbiota and especially components of the cell wall of Gram(-) bacteria: lipopolysaccharides (LPS). We have recently shown that LPS enhance glucagon-like peptide 1 (GLP-1) plasma levels, a hormone which is known to stimulate insulin secretion. Dietary lipids are detected by the gustatory system in rodents and humans. Two candidate lipid-receptors are found in taste buds: CD36, which is involved in the fat taste as shown by studies conducted in our laboratory, and GPR120. Our results show that GPR120 is not directly involved in the gustatory detection of lipids in mice, but could rather be involved in the modulation of the sensitivity for fat. When this gustatory system works properly, food choices can meet the organism’s energy needs. Besides, the pleasure brought by the consumption of palatable foods is satisfied. As part of a multi-year programme, survey campaigns have been carried out on both slopes of the Petit-Saint-Bernard pass (2188 m, western Alps), at an altitude of between 750 and 3000 m. The method employed abandons ground-based surveying in favour of multiple manual surveys carried out on selected topographic contexts down to the base of the Holocene fill.
