Isolation of endothelin A receptor from bovine lungs.
1995
We isolated endothelin receptor A (ET A ) from bovine lungs in a single-step purification procedure using antibodies raised against synthetic peptides that correspond to extra- and intracellular domains of the rat bradykinin receptor. Two receptor species of 55 and 35 kDa were isolated and subjected to N-terminal microsequencing. The difference between the observed and expected molecular weight species suggests that bovine ET A receptor is glycosylated.

Co-administration of pentoxifylline and thiopental causes death by acute pulmonary oedema in rats
2006
Background and purpose: Pentoxifylline exhibits rheological properties that improve microvascular flow and it is widely used in vascular perfusion disorders. It also exhibits marked anti-inflammatory properties by inhibiting tumour necrosis factor a production. Thiopental is one of the most widely used drugs for rapid induction of anaesthesia. During experimental studies on the treatment of acute pancreatitis, we observed that when pentoxifylline was administered after anaesthesia with thiopental, most of the rats exhibited dyspnea, signs of pulmonary oedema and died. The aim of the work described here was to investigate the cause of the unexpected toxic effect of the combined treatment wit…
In vivo antioxidant treatment protects against bleomycin-induced lung damage in rats
2003
1. This study examines the activity of the antioxidant N-acetylcysteine on bleomycin-induced pulmonary fibrosis in rats with emphasis on the early inflammatory phase. 2. Rats receiving N-acetylcysteine (300 mg kg(-1) day(-1), intraperitoneal) had less augmented lung wet weight, and lower levels of proteins, lactate dehydrogenase, neutrophil and macrophage counts in bronchoalveolar lavage fluid and lung myeloperoxidase activity with a betterment of histological score at 3 days postbleomycin. 3. A diminished lung GSH/GSSG ratio and augmented lipid hydroperoxides were observed 3 days postbleomycin. These changes were attenuated by N-acetylcysteine. Alveolar macrophages from bleomycin-exposed r…
Fatal Tumour Lysis Syndrome Induced by Brigatinib in a Lung Adenocarcinoma Patient Treated With Sequential ALK Inhibitors: A Case Report
2021
Tumour lysis syndrome (TLS) represents a group of fatal metabolic derangements resulting from the rapid breakdown of tumour cells. TLS typically occurs soon after the administration of chemotherapy in haematologic malignancies but is rarely observed in solid tumours. Here, we report a case of brigatinib-induced TLS after treatment with sequential anaplastic lymphoma kinase (ALK) inhibitors in a patient with advanced ALK-rearranged lung adenocarcinoma. The patient was treated sequentially with crizotinib, alectinib, and ensartinib. High-throughput molecular profiling after disease progression indicated that brigatinib may overcome ALK resistance mutations, so the patient was administered bri…
Oxidative stress and pulmonary inflammation: pharmacological intervention with antioxidants.
1999
Reactive oxygen and nitrogen species are generated by several inflammatory and structural cells of the airways. These oxidant species may have important effects on different lung cells as regulators of signal transduction, activators of key transcription factors, and modulators of gene expression and apoptosis. Thus, an increased oxidative stress accompanied by reduced endogenous antioxidant defences may have a role in the pathogenesis of a number of inflammatory pulmonary diseases including asthma. Although antioxidant drugs could play a useful role in the therapy of inflammatory lung diseases, their clinical impact is relatively modest at present. Rigorous clinical investigation with the …
Effect of Age on Systemic Exposure and Haematological Toxicity of Carboplatin in Advanced Non-Small Cell Lung Cancer Patients
2011
The aim of this study was to evaluate systemic exposure to carboplatin and its haematological toxicity in patients with advanced non-small cell lung cancer both older and younger than 70 years when the target area under the curve (AUC) in elderly patients was reduced by 20%. For this purpose, a population pharmacokinetic model was developed and the haema- tological toxicity of the drug was assessed. A total of 33 patients received carboplatin on day 1 and gemcitabine (1250 mg ⁄ m 2 ) on days 1 and 8. This schedule was repeated every 21 days. The Calvert-Crokcoft-Gault formula was employed to calculate a dose of carboplatin with a target AUC of 5 mg ⁄ min. ⁄ mL in patients under 70 years and…
Effect of quantized conductivity on the anomalous photon emission radiated from atomic-size point contacts
2019
We observe anomalous visible to near-infrared electromagnetic radiation emitted from electrically driven atomic-size point contacts. We show that the number of photons released strongly depends on the quantized conductance steps of the contact. Counter-intuitively, the light intensity features an exponential decay dependence with the injected electrical power. We propose an analytical model for the light emission considering an out-of-equilibrium electron distribution. We treat photon emission as bremsstrahlung process resulting from hot electrons colliding with the metal boundary and a find qualitative accord with the experimental data.
First operation of the KATRIN experiment with tritium
2020
AbstractThe determination of the neutrino mass is one of the major challenges in astroparticle physics today. Direct neutrino mass experiments, based solely on the kinematics of $$\upbeta $$β-decay, provide a largely model-independent probe to the neutrino mass scale. The Karlsruhe Tritium Neutrino (KATRIN) experiment is designed to directly measure the effective electron antineutrino mass with a sensitivity of $$0.2\hbox { eV}$$0.2eV ($$90\%$$90% CL). In this work we report on the first operation of KATRIN with tritium which took place in 2018. During this commissioning phase of the tritium circulation system, excellent agreement of the theoretical prediction with the recorded spectra was …
