Identification of the VvFLS2 grapevine flagellin receptor by a functional genomics strategy
2013
National audience; Grapevine (Vitis vinifera) is a crop of high agronomic interest subject to many destructive diseases. Pattern recognition receptors (PRRs) mediate detection of potential pathogens via the perception of pathogen associated molecular patterns (PAMPs), which is important for plant disease resistance. A well studied PRR is the Arabidopsis leucine-rich repeat receptor kinase FLS2 that recognizes bacterial flagellin (or its minimal motif flg22). Flagellin perception plays an important role in restricting bacterial invasion into plant leaves. Until now, functional FLS2 orthologues have been characterized in Nicotiana benthamiana, rice and tomato. Despite its economical importanc…

The sweet taste receptor is a heterodimer composed of two subunits called T1R2 and T1R3 whereas the T1R1 and the T1R3 subunits form a heterodimeric receptor for umami taste (the savory taste of monosodium glutamate). Each subunit belongs to the class C of G protein-coupled receptors (GPCRs) and is constituted by a large extracellular Nterminal domain (NTD) linked to the transmembrane domain by a cysteine-rich region. The NTD is composed of two lobes separated by a cleft in which ligands bind. T1R1- and T1R2-NTDs are able to bind sweeteners and umami compounds respectively and undergo ligand-dependent conformational changes (Zhang et al., 2008 ; Nie et al., 2005). However, the relative contr…

Abnormal peripubertal development of the rat mammary gland following exposure in utero and during lactation to a mixture of genistein and the food co…
2011
The impact of early exposure to endocrine disruptor mixtures on mammary gland development is poorly known. Here, we identify the effects of a conception to weaning exposure of rats to the phytoestrogen genistein (G) and/or the antiandrogen vinclozolin (V) at 1 mg/kg-d, alone or in association. Using several approaches, we found that G- and GV-exposed rats displayed significantly greater epithelial branching and proliferation, wider terminal end buds than controls at PND35, as well as ductal hyperplasia and periductal fibrosis. Focal branching defects were present in V-exposed rats. An increased ER and AR expression was observed in G- and CV- as compared to V-exposed rats at PND35. Surprisin…
Structure-function analysis of peroxisomal ATP-binding cassette transporters using chimeric dimers
2014
Background: Peroxisomal ABC transporters are predicted to function as homodimers in mammals. [br/] Results: ABCD1 interacts with ABCD2. Chimeric proteins mimicking full-length dimers represent novel tools for functional study. Artificial homodimers and heterodimers are functional. [br/] Conclusion: Interchangeability between ABCD1 and ABCD2 is confirmed, but PUFA transport depends on ABCD2. [br/] Significance: For the first time, heterodimers in mammals are proven to be functional.[br/] ABCD1 and ABCD2 are two closely related ATP-binding cassette half-transporters predicted to homodimerize and form peroxisomal importers for fatty acyl-CoAs. Available evidence has shown that ABCD1 and ABCD2 …
