Differential Proteomics Based on 2D-Difference In-Gel Electrophoresis and Tandem Mass Spectrometry for the Elucidation of Biological Processes in Ant…

2017

Proteomics based on 2D-Difference In Gel Electrophoresis (2D-DIGE) coupled with mass spectrometry (MS) procedures can be considered a “gold standard” to determine quantitatively and comparatively protein abundances in cell extracts from different biological sources/conditions according to a gel-based approach. In particular, 2D-DIGE is used for protein specie separation, detection, and relative quantification, whenever tandem MS is used to obtain peptide sequence information that is managed according to bioinformatic procedures to identify the differentially represented protein species. The proteomic results consist of a dynamic portray of over- and down-represented protein species that…

Analytical characterisation of the biodeterioration of diterpenoid labdanic varnishes used in pictorial techniques: Sandarac and Manila copal

2014

Abstract The deterioration produced by fungal and bacterial growth on the sandarac and Manila copal, diterpenoid varnishes traditionally used as art materials, was evaluated by gas chromatography–mass spectrometry (GC–MS), and pyrolysis–gas chromatography–mass spectrometry (Py–GC–MS). Test specimens, coated with a solidified thin layer of the studied varnishes, were inoculated and incubated with selected fungi and bacteria obtained from collections and oil paintings affected by biodeterioration, and analysed by the above mentioned techniques. Significant changes were detected, showing evidence of deterioration processes caused or favoured by some of the studied microorganisms.

The shell matrix of the european thorny oyster, Spondylus gaederopus: microstructural and molecular characterization.

2020

17 pages; International audience; Molluscs, the largest marine phylum, display extraordinary shell diversity and sophisticated biomineral architectures. However, mineral-associated biomolecules involved in biomineralization are still poorly characterised. We report the first comprehensive structural and biomolecular study of Spondylus gaederopus, a pectinoid bivalve with a peculiar shell texture. Used since prehistoric times, this is the best-known shell of Europe’s cultural heritage. We find that Spondylus microstructure is very poor in mineral-bound organics, which are mostly intercrystalline and concentrated at the interface between structural layers.Using high-resolution liquid chromatog…

A metabolomics cell-based approach for anticipating and investigating drug-induced liver injury

2016

AbstractIn preclinical stages of drug development, anticipating potential adverse drug effects such as toxicity is an important issue for both saving resources and preventing public health risks. Current in vitro cytotoxicity tests are restricted by their predictive potential and their ability to provide mechanistic information. This study aimed to develop a metabolomic mass spectrometry-based approach for the detection and classification of drug-induced hepatotoxicity. To this end, the metabolite profiles of human derived hepatic cells (i.e., HepG2) exposed to different well-known hepatotoxic compounds acting through different mechanisms (i.e., oxidative stress, steatosis, phospholipidosis…

The effect of Holder pasteurization on the lipid and metabolite composition of human milk

2022

Human milk (HM) is the gold standard for newborn nutrition. When own mother's milk is not sufficiently available, pasteurized donor human milk becomes a valuable alternative. In this study we analyzed the impact of Holder pasteurization (HoP) on the metabolic and lipidomic composition of HM. Metabolomic and lipidomic profiles of twelve paired HM samples were analysed before and after HoP by liquid chromatography-mass spectrometry (MS) and gas chromatography-MS. Lipidomic analysis enabled the annotation of 786 features in HM out of which 289 were significantly altered upon pasteurization. Fatty acid analysis showed a significant decrease of 22 out of 29 detectable fatty acids. The observed c…

Metabolomic Diversity of Human Milk Cells over the Course of Lactation—A Preliminary Study

2023

Human milk (HM) is a complex biofluid containing a wide cell variety including epithelial cells and leukocytes. However, the cellular compositions and their phenotypic properties over the course of lactation are poorly understood. The aim of this preliminary study was to characterize the cellular metabolome of HM over the course of lactation. Cells were isolated via centrifugation and the cellular fraction was characterized via cytomorphology and immunocytochemical staining. Cell metabolites were extracted and analyzed using ultra-performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry (UPLC–QqTOF-MS) in the positive and negative electrospray ionization mode…

Organic residues in lake and drinking water from the lagoa de Juturnaíba (região dos lagos - RJ, Brazil)

1997

Various analytical methods for the determination of organic residues in water were applied to samples from the Lagoa de Juturnaíba in the Região dos Lagos, east of Rio de Janeiro. The concentrations of more than 60 pesticides and of different aminopolycarboxylic acids were determined using GC/MS detection, and a GC/MS screening procedure was applied to detect additional compounds. None of the investigated pesticide residues could be detected in the lake water at detection limits of less than 100 ng/L. EDTA was detected at a concentration level of 2 mg/L in the lake and in drinking water prepared from this lake. Traces of biphenyl were identified by GC/MS in the lake water, the source of whi…

Isolation and identification of intermediates from biodegradation of low chlorinated biphenyls (Delor-103).

2004

Abstract Microorganism Pseudomonas species P2 metabolizes polychlorinated biphenyls (PCBs) and biphenyl, producing the whole spectrum of intermediates, among them coloured intermediates, which are suitable for the monitoring of PCBs degradation by optical sensors. Knowledge of chemical structures and conditions of development of colour metabolites is necessary for application of optical analytical methods. The main goal of this work was the isolation and identification of intermediates from the biodegradation of the mixture of low chlorinated biphenyls (Delor-103), which is based on the solid phase extraction (SPE) of the whole mixture using LiChrolut EN cartridges, then silylation of the e…

Analytical strategies for organic food packaging contaminants

2016

In this review, we present current approaches in the analysis of food-packaging contaminants. Gas and liquid chromatography coupled to mass spectrometry detection have been widely used in the analysis of some relevant families of these compounds such as primary aromatic amines, bisphenol A, bisphenol A diglycidyl ether and related compounds, UV-ink photoinitiators, perfluorinated compounds, phthalates and non-intentionally added substances. Main applications for sample treatment and different types of food-contact material migration studies have been also discussed. Pressurized Liquid Extraction, Solid-Phase Microextraction, Focused Ultrasound Solid-Liquid Extraction and Quechers have been …
