This chapter outlines the generation and application of human papillomavirus type 33 (HPV33) pseudovirions. The method describes (1) the construction of vaccinia viruses recombinant for the major and minor HPV capsid proteins, L1 and L2, respectively; (2) the transfection of Cos7 cells with a marker plasmid replicating to high copy numbers; (3) the expression of L1 and L2 using the vaccinia virus expression system; (4) the extraction, purification, and analysis of HPV33 pseudovirions; and (5) their use in pseudoinfection assays. SARS-CoV-2 envelope protein topology in eukaryotic membranes

Coronavirus E protein is a small membrane protein found in the virus envelope. Different coronavirus E proteins share striking biochemical and functional similarities, but sequence conservation is limited. In this report, we studied the E protein topology from the new SARS-CoV-2 virus both in microsomal membranes and in mammalian cells. Experimental data reveal that E protein is a single-spanning membrane protein with the N-terminus being translocated across the membrane, while the C-terminus is exposed to the cytoplasmic side (Nt lum /Ct cyt ). Characterization of the autoantigen La (SS-B) as a dsRNA unwinding enzyme

During the analysis of the La (SS-B) autoantigen for catalytic activities an ATP-dependent double-stranded RNA unwinding activity was detected. Both native and recombinant La proteins from different species displayed this activity, which could be inhibited by monospecific anti-La antibodies. La protein was able to melt dsRNA substrates with either two 3'-overhangs or a single 3'- and a 5'-overhang. Double-stranded RNAs with two 5'-overhangs were not unwound, indicating that at least one 3'-overhang is required for unwinding. Formaldehyde and total aldehydes in indoor air of public environments by voltammetry

The proposed method involves active sampling, where a sampling pump is used to pull air through a solution and voltammetric analysis of the obtained solutions. No interferences have been observed. In addition, very little sample preparation is required. Analyses were performed in 19 indoor stations and one in outdoor. Measurements were carried out on University environments: Museum of Chemistry, Zoological Museum, libraries, laboratories, corridors, meeting rooms, photocopying room, machine shop and terrace. Formaldehyde concentrations in analyzed samples ranged from 2.6 to 85 μg m-3 (median = 32 μg m-3), while the sum of others aldehydes ranged from 2 to 25 μg m-3 (median =2.4 μg m-3). In …
Micro-determination of dithiocarbamates in pesticide formulations using voltammetry

Abstract The purpose of this work was to develop a reliable method for the micro-determination of dithiocarbamates (mancozeb, maneb, propineb, nabam, Na(CH3)2DTC, zineb, ziram, ferbam and thiram) in pesticide formulations for agriculture using adsorptive stripping voltammetry (AdSV). The accuracy of analytical method was valued by analyzing simulate formulation samples prepared by us with known amounts of active ingredients. In addition, the applicability of the AdSV procedure for the analysis of DTCs and EBDTCs in micro-samples was evaluated by estimating its recoveries from spiked commercial formulate samples. International audience; Russell Banks’s 1985 novel Continental Drift tells the intertwined stories of Bob Dubois, a white American man who decides to try his luck in Florida, and the Haitian Vanise Dorsinville, who is forced to leave her island with her son and her nephew to flee Jean-Claude Duvalier’s dictatorial presidency. In this paper, I study how, in the chapters dedicated to the Haitian diaspora, Banks chooses to adopt an ex-centric point of view to give a voice to those whose history and culture have largely been ignored by American society and literature.
