Inside a Shell—Organometallic Catalysis Inside Encapsulin Nanoreactors

2021

Abstract Compartmentalization of chemical reactions inside cells are a fundamental requirement for life. Encapsulins are self‐assembling protein‐based nanocompartments from the prokaryotic repertoire that present a highly attractive platform for intracellular compartmentalization of chemical reactions by design. Using single‐molecule Förster resonance energy transfer and 3D‐MINFLUX analysis, we analyze fluorescently labeled encapsulins on a single‐molecule basis. Furthermore, by equipping these capsules with a synthetic ruthenium catalyst via covalent attachment to a non‐native host protein, we are able to perform in vitro catalysis and go on to show that engineered encapsulins can be used …

Hypsodont Myomiminae (Gliridae, Rodentia) from five new localities in the Lower Miocene Tudela Formation (Bardenas Reales, Ebro Basin, Spain) and the…

2012

The hypsodont Myomiminae Daams, 1981 (Gliridae) from the lower part of the Tudela Formation (Ebro Basin) are described. Five localities (CH1, CA2, CA3, CA4 and CC1) of this formation contain remains of the hypsodont genus Armantomy de Bruijn, 1966 and CC1 has also yielded Praearmantomys de Bruijn, 1966. Despite the poor material, we can identify the taxa A. cf. bijmai, A. daamsi (de Visser in Álvarez-Sierra, Daams, Lacomba, López Martínez, Van Der Meulen, Sesé & De Visser, 1991), A. cf. parsani, A. cf. jasperi and cf. P.crusafonti, which are characteristic of the Agenian and Ramblian continental stages. These data allow identification of the Agenian-Ramblian boundary in the Tudela Formation…

Beta-amyloid monomers are neuroprotective

2009

The 42-aa-long β-amyloid protein—Aβ1-42—is thought to play a central role in the pathogenesis of Alzheimer's disease (AD) (Walsh and Selkoe, 2007). Data from AD brain (Shankar et al., 2008), transgenic APP (amyloid precursor protein)-overexpressing mice (Lesné et al., 2006), and neuronal cultures treated with synthetic Aβ peptides (Lambert et al., 1998) indicate that self-association of Aβ1-42monomers into soluble oligomers is required for neurotoxicity. The function of monomeric Aβ1-42is unknown. The evidence that Aβ1-42is present in the brain and CSF of normal individuals suggests that the peptide is physiologically active (Shoji, 2002). Here we show that synthetic Aβ1-42monomers support …

Functional characterization of human nucleosome assembly protein-2 (NAP1L4) suggests a role as a histone chaperone.

1997

Abstract Histones are thought to play a key role in regulating gene expression at the level of DNA packaging. Recent evidence suggests that transcriptional activation requires competition of transcription factors with histones for binding to regulatory regions and that there may be several mechanisms by which this is achieved. We have characterized a human nucleosome assembly protein, NAP-2, previously identified by positional cloning at 11p15.5, a region implicated in several disease processes including Wilms tumor (WT) etiology. The deduced amino acid sequence of NAP-2 indicates that it encodes a protein with a potential nuclear localization motif and two clusters of highly acidic residue…

Determining a healthy reference range and factors potentially influencing PRO-C3 – A biomarker of liver fibrosis

2021

Background & Aims Progressive fibrosis has been identified as the major predictor of mortality in patients with non-alcoholic fatty liver disease (NAFLD). Several biomarkers are currently being evaluated for their ability to substitute the liver biopsy as the reference standard. Recent clinical studies in NAFLD/NASH patients support the utility of PRO-C3, a marker of type III collagen formation, as a marker for the degree of fibrosis, disease activity, and effect of treatment. Here we establish the healthy reference range, optimal sample handling conditions for both short- and long-term serum storage, and robustness for the PRO-C3 assay. Methods PRO-C3 was measured in 269 healthy volunteers…

Structural, Spectroscopic and Electrical Features of Undoped and Mn Doped LiTi2(PO4)3

2010

The study of the ionic conducting material LiTi2(PO 4)3 and of its Mn-substituted derivate reveals that the Mn distribution is strictly related to the synthetic method. The results of the structural refinement of X-ray and neutron (ToF) powder diffraction data and of XPS analysis demonstrate that Mn2+ ions are located on the lithium octahedral site, while Mn3+ and Mn4+ ions occupy the titanium ones. The Mn2+ amount on the lithium site seems to be the main factor responsible for the conductivity decrease observed in doped samples. The EPR spectra evidence clustering effects of Mn on both Li and Ti sites and the presence of more insulated Mn2+ ions. The effect of the major Mn amount on Ti sit…
