Simple Summary Hyaluronan is a main glycosaminoglycan in extracellular matrix with an important role in breast cancer progression. Alterations in its synthesis and size may affect tu-mour growth and metastasis. Triple-negative breast cancer (TNBC) is a clinically aggressive form of breast cancer that is unresponsive to endocrine agents or trastuzumab. TNBC accounts for ~10-20% of all breast cancer cases and represents the form with the poorest prognosis. AbstractNineteen pyrrolo[1,2-h][1,7]naphthyridinones and pyrido[2,3-c]pyrrolo[1,2-a]azepinones were synthesized as new tricyclic systems in which the pyridine ring is annelated to the 6,7-dihydroindolizin-8(5H)-one and 5,6,7,8-tetrahydro-9H-pyrrole[1,2-a]azepine-9-one moieties to obtain potential photosensitizing agents. They were tested for their photoantiproliferative activity on a triple-negative breast cancer cell line, MDA-MB-231, in the dark and under UVA light (2.0 J/cm2). Nanodaļiņas tiek uzskatītas par potenciālām transporta molekulām, kuras varētu nogādāt pretvēža zāles tieši uz audzēju šūnām. Bakalaura darba mērķis bija analizēt vai notiek nanodaļiņu pārnese uz audzēja šūnām mezinhimālo cilmes šūnu un MDA-MB-231 šūnu 3D kopkultūru modelī. Abas šūnu kultūras tika kultivēts S10 barotnē. Šūnu kultūru diferenciēšanai tika izvēlēts CD90 marķieris. MCŠ tika iezīmētas ar QD655 un pēc 6h inkubācijas tika veikta plūsmas citometrijas analīze. 3D kopkultūras modelis tika izveidots izmantojot PolyHEMA pārklājumu. QD pārnese uz vēža šūnām tika analizēta ar plūsmas citometriju pēc 48h kultivēšanas 3D kopkultūras modelī. 97% MCŠ 2D kultūrā pēc 6h inkubācijas uzņem QD655…
Vaccinum ģints ogu ekstraktu pretvēža aktivitātes pētīšana krūts vēža šūnu līnijās

Mūsdienās ar vien vairāk tiek meklēti jauni pretvēža līdzekļi, izmantojot augu valsts produktus. Maģistra darba mērķis bija noskaidrot, vai Vaccinium ģints ogu ekstrakti dažādās koncentrācijās ietekmē primārā krūts vēža MCF-7, metastātiskā krūts vēža MDA-MB-231 šūnas un ādas mezenhimālo cilmes šūnu D6 dzīvotspēju. Pētījumā tika izmantoti pieci ogu ekstrakti – mellene, krūmmellene, lielogu dzērvene, purva dzērvene un brūklene. Šūnām pievienoja ekstraktus koncentrācijās no 0,0016 mg/ml līdz 1 mg/ml un ar kolorimetrisku metodi (CCK-8) un mikroskopiju noteica šūnu dzīvotspēju un IC50. Study of the role of particle-particle dipole interaction in dielectrophoretic devices for biomarkers identification

A three dimensional Coupled Monte Carlo-Poisson method has been used to evaluate the impact of particle-particle dipole interactions in the equilibrium distribution of a system of uncharged polarizable particles suspended in a static liquid medium under the action of an oscillating non-uniform electric field generated by polynomial electrodes. We compare the simulated distributions with experimental ones both for micro- (MDA-MB-231 breast tumor cells) and nano-(multiwall carbon nanotubes) particles. In both cases the equilibrium distributions near the electrodes are dominated by dipole interactions which locally enhance the DEP effect and promote long particles chains.
Insight on pyrimido[5,4-g]indolizine and pyrimido[4,5-c]pyrrolo[1,2-a]azepine systems as promising photosensitizers on malignant cells

Searching for new small molecules as photosensitizing agents, we have developed a class of twenty-five pyrimido[5,4-g]indolizine and pyrimido[4,5-c]pyrrolo[1,2-a]azepines with a good substitution pattern defining a versatile synthetic pathway to approach the title ring system. All compounds were evaluated for their photocytotoxicity on a triple negative human breast cancer cell line (MDA-MB-231) in the dark and under UVA light (2.0 J/cm2). The most effective compounds exhibited a photoantiproliferative activity with IC50 values up to nanomolar ranges.
