Diferenciācijas indukcijas izpēte pēc BRAF un mTOR inhibitoru apstrādes BRAF mutētās ādas melanomas šūnās

Darba mērķis bija padziļināti izpētīt BRAF inhibīcijas un sekojoši kombinētās BRAF un mTOR inhiibīcijas ietkemi uz metastātiskās BRAF mutēās melanomas šūnu līnijas SKMel28 šūnu ciklu, dzīvotspēju un inducēto atgriezensiko epitelilālo diferenciāciju. Ar advancētām mikroskopijas un šūnu analīzes metodēm tika apskatīti šos procesus iespējami ietkmējošie mehānismi. BRAF inhibīcija ar Vemurafenib rezultējās melanomas šūnu epiteliālā diferenciācijā un G1 arestā. Kombinētā BRAF un mTOR inhibīcija šūnās pagarināja šūnu diferencētā fenotipa stadiju, kas norāda uz iespējamu PI3K ceļa iesaisti SKMel28 šūnu augšanas un proliferācijas procesos. Translation of HTT mRNA with expanded CAG repeats is regulated by the MID1-PP2A protein complex.

Expansion of CAG repeats is a common feature of various neurodegenerative disorders, including Huntington's disease. Here we show that expanded CAG repeats bind to a translation regulatory protein complex containing MID1, protein phosphatase 2A and 40S ribosomal S6 kinase. Binding of the MID1-protein phosphatase 2A protein complex increases with CAG repeat size and stimulates translation of the CAG repeat expansion containing messenger RNA in a MID1-, protein phosphatase 2A- and mammalian target of rapamycin-dependent manner. Nustatyta, kad mokinių įsitikinimai apie savo gebėjimus, susidomėjimas lietuvių kalba, tėvų lūkesčiai bei 2-os klasės pasiekimai tarpusavyje siejasi teigiamais ryšiais. Glycogen synthase kinase 3 (GSK-3) is a serine/threonine (S/T) protein kinase. Although GSK-3 originally was identified to have functions in regulation of glycogen synthase, it was subsequently determined to have roles in multiple normal biochemical processes as well as various disease conditions. GSK-3 is sometimes referred to as a moonlighting protein due to the multiple substrates and processes which it controls. Frequently, when GSK-3 phosphorylates proteins, they are targeted for degradation. The electro-optical properties of most semiconductors and insulators of technological interest are dominated by the presence of electron-hole quasi-particles, called excitons. The manipulation of excitons in dielectrics has recently received great attention, with possible applications in different fields including optoelectronics and photonics. Here, we apply attosecond transient reflection spectroscopy in a sequential two-foci geometry and observe sub-femtosecond dynamics of a core-level exciton in bulk MgF2 single crystals. One-dimensional pion, kaon, and proton femtoscopy in Pb-Pb collisions at √sNN = 2.76 TeV

The size of the particle emission region in high-energy collisions can be deduced using the femtoscopic correlations of particle pairs at low relative momentum. Such correlations arise due to quantum statistics and Coulomb and strong final state interactions. In this paper, results are presented from femtoscopic analyses of π±π±, K±K±, K0 SK0 S, pp, and pp correlations from Pb-Pb collisions at √sNN = 2.76 TeV by the ALICE experiment at the LHC. One-dimensional radii of the system are extracted from correlation functions in terms of the invariant momentum difference of the pair. Synthesis and Antitumor Evaluation of Menthone-Derived Pyrimidine-Urea Compounds as Potential PI3K/Akt/mTOR Signaling Pathway Inhibitor.

A series of novel menthone derivatives bearing pyrimidine and urea moieties was designed and synthesized to explore more potent natural product-derived antitumor agents. The structures of the target compounds were confirmed by FTIR, NMR, and HRMS. The in vitro antitumor activity was tested by standard methyl thiazolytetrazolium assay and showed that 4i, 4g, 4s, and 4m are the best compounds with IC50 values of 6.04 ± 0.62µM, 3.21 ± 0.67µM, 19.09 ± 0.49µM, and 18.68 ± 1.53µM, against Hela, MGC-803, MCF-7, and A549, respectively.
