Carbon based nanomaterials for the detection of narrow therapeutic index pharmaceuticals
2020
Precise detection of important pharmaceuticals with narrow therapeutic index (NTI) is very critical as there is a small window between their effective dose and the doses at which the adverse reactions are very likely to appear. Regarding the fact that various pharmacokinetics will be plausible while considering pharmacogenetic factors and also differences between generic and brand name drugs, accurate detection of NTI will be more important. Current routine analytical techniques suffer from many drawbacks while using novel biosensors can bring up many advantages including fast detection, accuracy, low cost with simple and repeatable measurements. Recently the well-known carbon Nano-allotrop…
Mass Transport Analysis of Bicarbonate Buffer: Effect of the CO2–H2CO3 Hydration–Dehydration Kinetics in the Fluid Boundary Layer and the Apparent Ef…
2019
The main buffering system influencing ionizable drug dissolution in the human intestinal fluid is bicarbonate-based; however, it is rarely used in routine pharmaceutical practice due to the volatility of dissolved CO2. The typical pharmaceutical buffers used fail to capture the unique aspects of the hydration-dehydration kinetics of the bicarbonate-CO2 system. In particular, CO2 is involved in a reversible interconversion with carbonic acid (H2CO3), which is the actual conjugate acid of the system, as follows CO2 + H2O ⇌ H2CO3. In contrast to ionization reactions, this interconversion does not equilibrate very rapidly compared to the diffusional processes through a typical fluid diffusion b…
Highly Fluorescent and Water-Soluble Diketopyrrolopyrrole Dyes for Bioconjugation
2015
International audience; The preparation of highly water-soluble and strongly fluorescent diketopyrrolopyrrole (DPP) dyes using an unusual taurine-like sulfonated linker has been achieved. Exchanging a phenyl for a thienyl substituent shifts the emission wavelength to near λ=600 nm. The free carboxylic acid group present in these new derivatives was readily activated and the dyes were subsequently covalently linked to a model protein (bovine serum albumin; BSA). The bioconjugates were characterized by electronic absorption, fluorescence spectroscopy and MALDI-TOF mass spectrometry, thus enabling precise determination of the labeling density (ratio DPP/BSA about 3 to 8). Outstanding values of…
Peracarid Crustacea Inhabiting Aegagropylae of the Red Alga Rytiphloea Tinctoria (Clemente) C. Ag. in the Stagnone Sound, Western Sicily, Italy
1993
The peracarid Crustacea belonging to Cumacea, Tanaidacca, Isopoda and Amphipoda, inhabiting aegagropylae of Rytiphloea tinctoria were studied throughout a year in the Stagnone Sound (western Sicily, Italy). The dominant groups, tanaids (61.7%) and amphipods (32.8%), were represented by 6 and 28 species, and showed complementary trends during the 12 months, reaching their maximum of abundance in spring and autumn, respectively. Tanaids were mainly represented by Apseudes intermedius Hansen, 1895 and Parapseudes latifrons (Grube, 1864), and amphipods by Elasmopus pocillimanus (Bate, 1862), Leptocheirus guttatus (Grube, 1864), Lysianassa longicornis Lucas, 1849 and Maera inaequipes (A. Costa, …
Cytotoxicity of oleanolic and ursolic acid derivatives toward hepatocellular carcinoma and evaluation of NF-κB involvement.
2019
Oleanolic and ursolic acids are two ubiquitous isomeric triterpene phytochemicals known for their anticancer activity. A set of derivatives of the two compounds with a modified oxidation state and lipophylicity at C-3 and C-28 positions, were prepared and tested as anticancer agents versus the lines HepG2, Hep3B and HA22T/VGH of hepatocarcinoma, a strongly aggressive tumor that is not responsive toward the standard therapies. New derivatives containing a three carbons side chain on the C-3 position were synthetized in both stereoisomeric forms by the Barbier-Grignard procedure and three of them were found to be active toward all of the three targets. The implication of the transcriptional n…
Doxorubicin induces wide-spread transcriptional changes in the myocardium of hearts distinguishing between mice with preserved and impaired cardiac f…
2021
Abstract Aims Doxorubicin (DOX) is an important drug for the treatment of various tumor entities. However, the occurrence of heart failure limits its application. This study investigated differential gene expression profiles in the left and right ventricles of DOX treated mice with either preserved or impaired myocardial function. We provide new mechanistic insights into the pathophysiology of DOX-induced heart failure and have discovered pathways that counteract DOX-induced cardiotoxicity. Main methods We used in total 48 male mice and applied a chronic low dose DOX administration (5 mg/kg per injection, in total 20 mg/kg over 4 weeks) to induce heart failure. Echocardiographic parameters …
Cardioprotective effects of phytopigments via multiple signaling pathways.
2021
Abstract Background Cardiovascular diseases (CVDs) are among the deadliest non-communicable diseases, and millions of dollars are spent every year to combat CVDs. Unfortunately, the multifactorial etiology of CVDs complicates the development of efficient therapeutics. Interestingly, phytopigments show significant pleiotropic cardioprotective effects both in vitro and in vivo. Purpose This review gives an overview of the cardioprotective effects of phytopigments based on in vitro and in vivo studies as well as clinical trials. Methods A literature-based survey was performed to collect the available data on cardioprotective activities of phytopigments via electronic search engines such as Pub…
New cytostatic agents obtained by molecular topology
1996
Abstract Four new cytostatic compounds have been obtained by computer aided selection based on Molecular Connectivity, a topological approach to molecular structural study. Three of them, namely tetracycline, doxycycline and piromidic acid are well known antibiotic agents, and the fourth, carminic acid, is an innocuous colorant. Although no previous report of activity was found, on the used cell lines, for any of the selected compounds, carminic acid may be considered a new lead one since no structure-related cytostatic molecules have been reported in the literature. These results are interesting for two reasons: First, new potential low-toxicity anticancer drugs show to be a possible thera…
Physicochemical stability of carmustine-containing medicinal products after reconstitution and after dilution to ready-to-administer infusion solutio…
2020
Introduction The aim of this study was to determine and compare the physicochemical stability of two carmustine-containing medicinal products licensed and marketed in Europe as Carmustin Obvius (Medac GmbH) and Carmubris (Tillomed Pharma GmbH). Reconstituted stock solutions and diluted ready-to-administer infusion solutions of the two products were investigated. Methods Reconstituted carmustine stock solutions (3.3 mg/mL) and ready-to-administer infusion solutions (0.2 mg/mL, 1.0 mg/mL) prepared in prefilled 5% glucose injection solution PP/PE bags were stored at 22°C or 2–8°C over a maximum period of 66 hours protected from light. Samples were taken immediately after reconstitution or dilu…
Modulating carbohydrate-based hydrogels as viscoelastic lubricant substitute for articular cartilages
2017
Viscosupplementation is a therapeutic approach for osteoarthritis treatment, where the synovial fluid, the natural lubricant of the joints, is replaced by viscoelastic solutions with rheological properties comparable or better than the starting material. This study presents the development of an innovative platform for viscosupplementation, based on the optimization of polysaccharide-based colloidal hydrogel, aiming to reduce on-site enzyme degradation and enhance the possibility of hyaluronic acid substitution with alternative biomaterials. Catanionic vesicles are proposed as physical crosslinker that can guarantee the formation of a 'soft', tunable network, offering a dual-therapeutic app…
