Hyperpigmentation of hard palate induced by chloroquine therapy

The antimalarials are one of the most commonly prescribed drugs for conditions such as lupus erythematosus and rheumatoid arthritis, and the side effects, though infrequent, are well known. The antimalarial agent chloroquine diphosphate usually causes pigmentary changes in the oral mucosa characterized by a bluish-grey to black discolorations mainly in the hard palate. Considering only the hard palate hyperpigmentation caused by chloroquine, to the best of our knowledge, only 13 cases have been reported in the English language literature. El primer capítulo se centra principalmente en el ámbito de la química médica, donde se discute el diseño, la síntesis y la evaluación biológica de cuatro familias de compuestos con potencial actividad antimalárica. Para el desarrollo de este trabajo se establece un convenio de colaboración con la empresa farmacéutica GSK, la cual lleva a cabo la evaluación biológica de todos los compuestos sintetizados. Herein we report the synthesis of a set of constrained peptidomimetics endowed with an electrophilic vinyl ester warhead and structurally related to a previously identified lead compound, a potent and irreversible inhibitor of falcipain-2 (FP-2). FP-2 is the main hemoglobinase of the malaria parasite P. falciparum. The new compounds were evaluated for their inhibition against FP-2, and the results were rationalized on the basis of docking experiments.
