No evidence of beneficial effects of plasmapheresis in natalizumab-associated PML

2017

Objective:To examine retrospectively the effects of plasmapheresis (PLEX) on the survival and clinical outcomes of patients with multiple sclerosis (MS) and natalizumab (NTZ)–associated progressive multifocal leukoencephalopathy (PML).Methods:The medical literature was searched for the terms natalizumab and progressive multifocal leukoencephalopathy. A total of 193 international and 34 Italian NTZ-PML cases were included. Clinical outcome was determined by comparing the patients' clinical status at PML diagnosis with status after PML resolution. The effects on survival and clinical outcome of PLEX, sex, age, country, pre-PML Expanded Disability Status Scale score, NTZ infusion number, prior…

Acquired BRAF inhibitor resistance: A multicenter meta-analysis of the spectrum and frequencies, clinical behaviour, and phenotypic associations of r…

2015

BackgroundAcquired resistance to BRAF inhibitors (BRAFi) is a near-universal phenomenon caused by numerous genetic and non-genetic alterations. In this study, we evaluated the spectrum, onset, pattern of progression, and subsequent clinical outcomes associated with specific mechanisms of resistance.MethodsWe compiled clinical and genetic data from 100 patients with 132 tissue samples obtained at progression on BRAFi therapy from 3 large, previously published studies of BRAFi resistance. These samples were subjected to whole-exome sequencing and/or polymerase chain reaction-based genetic testing.ResultsAmong 132 samples, putative resistance mechanisms were identified in 58%, including NRAS o…

Genomic profile of breast cancer: costeffectiveness analysis from the Spanish National Healthcare System perspective.

2014

Background: Costeffectiveness analysis of MammaPrint® (70-gene signature) in the diagnosis of early breast cancer as a prognosis assay to study the risk of tumor recurrence to administer adjuvant chemotherapy. Methods: Markov model assuming a cohort of 60-year-old women with breast cancer. Treatment costs and effects were assessed by comparing the 5-year, 10-year and lifetime risk of recurrence using Adjuvant! Online® (online algorithm), 70-gene signature or Oncotype DX® (21-gene assay). Results: 70-gene signature showed a life expectancy of 23.55 years at lifetime. Life expectancy was lower for 21-gene assay and online algorithm, with associated quality-adjusted life year gains up to 0.23 …

Ontology mapping specification in description logics for cooperative systems

2006

Ontology Views for Ontology Change Management

2014

International audience; In the literature, ontology change management systems (OCMS) are direct implementation of the concept of “change management” stated by reference (Klein, 2004). Ontology change management combines ontol- ogy evolution and versioning features to manage ontol- ogy changes and their impacts. Since 2007, many works have combined ontology evolution and versioning into ontology change management systems (OCMS). The evolution subject has been massively studied in these works. They especially addressed the consistence issue for the application of changes on the ontology. These proposals constituted a consequent background for ontology change management but they did not take i…

Analysis of technical criticalities for GIS modelling an Urban noise map

2017

This paper analyzes criticalities and strengthens of a procedure used to model the acoustic map of the vehicular traffic of an urban agglomeration. The research is based on a pilot project for the acoustic mapping of a portion of the city of Palermo (Italy). Simulations indicate that the acoustic climate was in line with expectations and with typical of large Italian cities. The most remarkable result was obtained by an increase in the number of reflections (from two to five), while the influence of the height of the building (from 9 to 18 meters) was negligible – on the order of a few points per thousand. Regarding the analysis conducted with the ‘Gden Method’, acoustic values do not diver…

A genetic algorithm for analyzing choice behavior with mixed decision strategies

2009

In the field of decision-making a fundamental problem is how to uncover people's choice behavior. While choices them- selves are often observable, our underlying decision strategies determining these choices are not entirely understood. Previous research defined a number of decision strategies and conjectured that people do not apply only one strategy but switch strategies during the decision process. To the best of our knowledge, empirical evidence for the latter conjecture is missing. This is why we monitored the purchase decisions 624 consumers shopping online. We study how many of the observed choices can be explained by the existing strategies in their pure form, how many decisions can…

Cloning of branched chain amino acid biosynthesis genes and assays of alpha-acetolactate synthase activities in Leuconostoc mesenteroides subsp. crem…

1999

Abstract A genomic library from Leuconostoc mesenteroides subsp. cremoris (Lmc) in Escherichia coli was screened for α-acetolactate synthase (ALS) activity using a phenotypic test detecting the production of acetolactate or related C 4 derivatives (diacetyl, acetoin or 2,3-butanediol) in the culture. Four recombinant E. coli clones, with plasmids containing overlapping DNA fragments and displaying anabolic ALS activity, were selected. This activity is encoded by an ilvB gene belonging to a putative operon which contains genes highly similar to the genes of the branched chain amino acid (BCAA) operon of Lactococcus lactis subsp. lactis. This putative BCAA operon is not functional as the ilvA…
