Recent genome-wide association studies have linked type-2 diabetes mellitus to a genomic region in chromosome 9p21 near the Ink4/Arf locus, which encodes tumor suppressors that are up-regulated in a variety of mammalian organs during aging. However, it is unclear whether the susceptibility to type-2 diabetes is associated with altered expression of the Ink4/Arf locus. Histone deacetylase inhibitors (HDACIs) are epigenetic compounds that have been recently considered for their promising anti-tumor activity. The aim of this PhD thesis was to elucidate and characterize the anti-tumor effect of the HDAC inhibitor ITF2357 (Givinostat) in melanoma and colon cancer cells that are characterized by oncogenic BRAF mutations. Interestingly, data reported in this thesis demonstrate that ITF2357 exerts a remarkable anti-tumor effect in melanoma cells by inducing a switch from a pro-survival autophagy to caspase-dependent apoptosis. Oncogenic BRAF mutations have been widely described in melanomas and promote tumour progression and chemoresistance. We previously provided evidence that the HDAC inhibitor ITF2357 (Givinostat) targets oncogenic BRAF in SK-MEL-28 and A375 melanoma cells. Here, we show that oncogenic BRAF localises to the nucleus of these cells, and the compound decreases BRAF levels in both the nuclear and cytosolic compartments. Although mutations in the tumour suppressor p53 gene are not equally frequent in melanomas compared to BRAF, the functional impairment of the p53 pathway may also contribute to melanoma development and aggressiveness. Il saggio descrive molte delle opere d'arte del XIV e XV secolo esposte in mostra (dipinti su tavola, sculture lignee, oggetti in argento con smalti, cofanetti in avorio e legno), di fattura locale, italiana, cipriota-egea, iberica, francese e tedesca, tutte provenienti dalla Sicilia, alcune legate alla famiglia Chiaromonte. The essay describes many of the 14th and 15th century works of art on display (paintings on wood, wooden sculptures, silver objects with enamels, ivory and wooden boxes), of local, Italian, Cypriot-egean, Iberian, French and German manufacture, all from Sicily, some related to the Chiaromonte family.

Melt Processed PCL/PEG Scaffold with Discrete Pore Size Gradient for Selective Cellular Infiltration

In order to develop scaffold able to mimic the natural gradient properties of tissues, biphasic and triphasic approaches were adopted. In this work, polycaprolactone/polyethylene glycol (PCL/PEG) scaffolds were prepared by using a combination of melt mixing and selective leaching without harmful solvents. The method permitted to develop three-layer scaffolds with high control of porosity and pore size. The mechanical properties were evaluated under physiological condition in order to simulate the real conditions of work. Co-culture of osteoblastic and fibroblastic mice cells were carried out in order to study the differential cellular permeation through the different pore size layers.

In the field of international jurisdiction, they strive to align the competence in couples? patrimony disputes to that in succession and in separation proceedings, or else to align the competence of the courts to the applicable law. These tendencies are clearly visible in the Regulations? provisions on choice of court agreements.
