Using the Hermite Regression Formula to Design a Neural Architecture with Automatic Learning of the “Hidden” Activation Functions

2000

The value of the output function gradient of a neural network, calculated in the training points, plays an essential role for its generalization capability. In this paper a feed forward neural architecture (αNet) that can learn the activation function of its hidden units during the training phase is presented. The automatic learning is obtained through the joint use of the Hermite regression formula and the CGD optimization algorithm with the Powell restart conditions. This technique leads to a smooth output function of αNet in the nearby of the training points, achieving an improvement of the generalization capability and the flexibility of the neural architecture. A Feature Rich Distance-Based Many-Objective Visualisable Test Problem Generator

2019

In optimiser analysis and design it is informative to visualise how a search point/population moves through the design space over time. Visualisable distance-based many-objective optimisation problems have been developed whose design space is in two-dimensions with arbitrarily many objective dimensions. Previous work has shown how disconnected Pareto sets may be formed, how problems can be projected to and from arbitrarily many design dimensions, and how dominance resistant regions of design space may be defined. Most recently, a test suite has been proposed using distances to lines rather than points. However, active use of visualisable problems has been limited. This may be because the ty…

Site-specific near-infrared fluorescent labelling of proteins on cysteine residues with meso -chloro-substituted heptamethine cyanine dyes

2018

International audience; Near-infrared (NIR) fluorescence imaging is a promising new medical imaging modality. Associated with a targeting molecule, NIR fluorophores can accumulate selectively in tissues of interest and become valuable tools for the diagnosis and therapy of various pathologies. To facilitate the design of targeted NIR imaging agents, it is important to identify simple and affordable fluorescent probes, allowing rapid labelling of biovectors such as proteins, ideally in a site-specific manner. Here, we demonstrate that heptamethine cyanine based fluorophores, such as IR-783, that contain a chloro-cyclohexyl moiety within their polymethine chain can react selectively, at neutr…

Synthesis of No-Carrier-Added 4-[18F]Fluorophenol from 4-Benzyloxyphenyl-(2-thienyl)iodonium Bromide

2011

4-[(18)F]Fluorophenol is a versatile synthon for the synthesis of more complex radiopharmaceuticals bearing a 4-[(18)F]fluorophenoxy moiety. In order to prepare 4-[(18)F]fluorophenol in no-carrier-added (n.c.a.) form only a nucleophilic labelling method starting from [(18)F]fluoride is suitable. In this paper a new, two step radiosynthesis starting from 4-benzyloxyphenyl-(2-thienyl)iodonium bromide and [(18)F]fluoride with subsequent deprotection is described, yielding n.c.a. [(18)F]fluorophenol in 34 to 36% radiochemical yield.

Chemosensors displaying pH controlled multistage fluorescence emission

1999

A large number of systems displaying pH controlled multistages of fluorescence emission intensity, have not been recognized from this point of view. In this work we present two paradigmatic cases: (i) polyamine receptors linked to a fluorophore unit, in which the signal is modulated by an intra-molecular pH dependent electron transfer process, (ii) phenols presenting adiabatic excited state proton transfer. In the first example the chemosensors are constituted by a receptor unit containing a polyaza moiety suitable for binding protons (but also metal ions or anions). The receptor is linked to a benzene or naphthalene unit that acts as a fluorophore. The fluorescence signal is dependent on t…

Azide and sulfonylazide functionalized fluorophores for the selective and sensitive detection of hydrogen sulfide

2015

[EN] Three fluorescent probes (1–3) for the selective and sensitive detection of hydrogen sulfide have been synthesized and characterized. Probe 1 is a coumarin derivative functionalized with an azide moiety whereas 2 contain the azide reactive group into a naphthalene fluorophore backbone. Probe 3 is composed also by a naphthalene fluorophore but, in this case, functionalized with a sulfonylazide reactive moiety. Probes 1 and 3 are non-fluorescent whereas 2 is weakly emissive in HEPES (10 mM, pH 7.4)–DMSO 99:1 (v/v). The emission behavior of the three probes was tested against selected anions, bio-thiols and oxidant molecules. Of all the chemical species tested, only HS− is able to induce …

A micellar multitasking device: sensing pH windows and gauging the lipophilicity of drugs with fluorescent signals.

2010

A multitasking fluorescent device can be obtained by forming micelles of Triton X-100, containing a lipophilic macrocyclic Cu(2+) complex and the coordinating fluorophore Coumarin 343 (C343), which features a COOH moiety. At low pH the two micellised components do not interact, and the fluorescence of Courmarin 343 (C343) is intense. At intermediate pH, C343 is deprotonated and coordinates to the Cu(2+) centre in its apical position, with fluorescence quenching. At higher pH the deprotonated C343 is displaced from Cu(2+) by the formation of an OH(-) complex, and the fluorescence is revived. This allows the system to carry out its first task as it behaves as an "on-off-on" fluorescent sensor…

A FRET-based probe for fluorescence sensing of sulfide/sulfite analytes, using a novel long-wavelength water-soluble 7-hydroxycoumarin as report…

2015

International audience; A FRET-based fluorescent probe for the detection of sulfide and/or sulfite in aqueous buffer, was constructed by connecting the quencher moiety nitrobenzofurazan (7-nitro-1,2,3-benzoxadiazole, NBD) to a water-soluble 3-(2-benzimidazolyl)-7-hydroxycoumarin carboxylic acid through a piperazine linker. This probe exhibits good selectivity and high sensitivity for sulfite and sulfide over cysteine and other potential analytes. Furthermore, this is one of the few examples of fluorogenic coumarins whose solubility in water is maintained upon its conversion into reaction-based fluorescent probes.
