Detection of anti-myeloperoxidase antibodies in the serum of patients with type 1 diabetes mellitus.

Anti-myeloperoxidase (anti-MPO) antibodies were detected in 34 of 88 (38%) patients with type 1 diabetes mellitus but in only 3 of 55 (5.7%) healthy subjects and in 4 of 20 patients with autoimmune disease. Specificity of anti-MPO antibodies was assessed by MPO inhibition studies. No relationship was found between the occurrence of anti-MPO and anti-thyroperoxidase antibodies. Levels of soluble intercellular adhesion molecule 1 (ICAM-1) were found to be higher in anti-MPO antibody-positive (n = 28, 508 +/- 126 ng/ml) than in anti-MPO antibody-negative (n = 58, 438 +/- 140 ng/ml: P < 0.05) patients. Actin in Allomyces arbuscula

International audience; A 42 kDa protein was isolated by affinity chromatography on DNAse I-Sepharose from 24-h-old mycelia of Allomyces arbuscula. It was identified as actin by immunoblots with monoclonal antibody probe against a chicken gizzard actin. This fungal actin has a pI of 5.9 and separates into two spots on two-dimensional polyacrylamide gel electrophoresis, suggesting its dual nature. It can polymerize into 8-10 nm filaments visualized by electron microscopy.

The cDNA for the full-length lectin from the marine sponge Geodia cydonium was cloned. Analysis of the deduced aa sequence revealed that this lectin belongs to the group of galectins. The full-length galectin, which was obtained also in a recombinant form, has an M(r) of 20,877; in the processed form it is a 15 kDa polypeptide. The enriched aggregation factor from G.cydonium also was determined to contain, besides minimal amounts of the galectin, a 140 kDa polypeptide which is involved in cell-cell adhesion. To study the sequential expression of insect epidermal cells during metamorphosis, a library of monoclonal antibodies (MABs) was prepared against the water-soluble proteins from preecdysial pupal cuticle of Tenebrio molitor. Six selected MABs recognizing only larval and pupal cuticular proteins (CPs) in immunoblot analysis were classified into three types. Type 1 recognized a 21.5 and a 22 kDa polypeptide, type 2, a 26 kDa polypeptide, and type 3, three polypeptides of 18.5, 19.5 and 21.5 kDa. The use of a novel monoclonal antibody (mAb) that reacts with (1,6)-beta-glucan has permitted the study of the different covalent linkages between glucan and mannoproteins in the cell wall of Candida albicans. The mAb JRR1 was originally raised by immunization with Zymolyase extracts from C. albicans cell walls, but it soon became apparent that it reacted with a (1,6)-beta-glucan epitope. By using this antibody, we show the existence of glucan-mannoprotein complexes between the (1,6)-beta-glucan epitope recognized by the antibody and cell wall mannoproteins. The topology of the (1,6)-beta-glucan in the cell wall of C. albicans has also been studied.

In this study we describe the site and moment of histospecific differentiation in developmental stages of the annelid Platynereis dumerilii by use of biochemical markers. The monoclonal antibody (mab) OI7 and uncloned hybridoma supernatants (pAb's) OI8, OI10, OI46 and OI69 recognize neural antigens that appear asynchronously during development. By an enzymatic test, acetylcholinesterase (AChE) was found specific for nervous tissue as well.
