Between 2008 and 2011, the head of 150 Euthynnus alletteratus (Osteichthyes: Scombridae) caught inshore off the southeastern Iberian coast (western Mediterranean Sea) were examined for parasites. Two monogeneans, four didymozoid trematodes and four copepods were found. Parasite abundance showed a positive relationship with the annual sea surface temperature, except for Pseudocycnus appendiculatus, but negative with the sea depth (Capsala manteri, Neonematobothrium cf. kawakawa and Caligus bonito). [Background] Monogenean flatworms are the main fish ectoparasites inflicting serious economic losses in aquaculture. The polyopisthocotylean Sparicotyle chrysophrii parasitizes the gills of gilthead sea bream (GSB, Sparus aurata) causing anaemia, lamellae fusion and sloughing of epithelial cells, with the consequent hypoxia, emaciation, lethargy and mortality. Next-generation sequencing (NGS) panels are used widely in clinical diagnostics to identify genetic causes of various monogenic disease groups including neurometabolic disorders and, more recently, lysosomal storage disorders (LSDs). Many new challenges have been introduced through these new technologies, both at the laboratory level and at the bioinformatics level, with consequences including new requirements for interpretation of results, and for genetic counseling. Congenital goitrous primary hypothyroidism in two German families caused by novel thyroid peroxidase (TPO) gene mutations.

Congenital hypothyroidism occurs with a prevalence of approximately 1:3 500. Defects in thyroid hormone synthesis which lead to goitrous hypothyroidism account for 10-15% of these cases. Several genetic defects have been characterized and mutations in the thyroid peroxidase (TPO) gene are the most common cause for dyshormonogenesis.So far, more than 80 mutations in the TPO gene have been described, resulting in a variable decrease in TPO bioactivity. Patients with diabetes caused by single-gene mutations generally exhibit an altered course of diabetes. Those with mutations of the glucokinase gene (GCK-MODY) show good metabolic control and low risk of cardiovascular complications despite paradoxically lowered high-density lipoprotein (HDL) cholesterol levels. Dactylogyrus (Monogenea) communities on the gills of roach in three lakes in Central Finland

SUMMARYDactylogyrus (Monogenea) species were studied on the gills of roach (Rutilus rutilus) from a total of 293 fish in 3 interconnected lakes in Central Finland. One of the lakes is eutrophic and polluted by a paper and pulp mill, one is eutrophic and one is oligotrophic. Nine Dactylogyrus species were found. The structure of the monogenean communities was analysed at the component and infracommunity level and the Dactylogyrus fauna of the lakes was compared. Dactylogyrus (Monogenea) infections on the gills of roach (Rutilus rutilus L.) experimentally exposed to pulp and paper mill effluent

Experimental exposure to bleached kraft pulp and paper mill effluent (BKME) at a concentration of 10% significantly reduced the abundance and mean number of species of Dactylogyrus on the gills of naturally infected roach, Rutilus rutilus, over a 3 week period. Seven Dactylogyrus species were recorded which differed widely in their susceptibility to the effluent. The experiment coincided with a natural spring peak in dactylogyrid infections on roach. BMKE exposure did not prevent parasite reproduction but post-larval abundance was significantly higher on control than effluent-exposed hosts.
