Polyoxypregnanes as safe, potent, and specific ABCB1-inhibitory pro-drugs to overcome multidrug resistance in cancer chemotherapy in vitro and in vivo

Multidrug resistance (MDR) mediated by ATP binding cassette subfamily B member 1 (ABCB1) is significantly hindering effective cancer chemotherapy. However, currently, no ABCB1-inhibitory drugs have been approved to treat MDR cancer clinically, mainly due to the inhibitor specificity, toxicity, and drug interactions. Here, we reported that three polyoxypregnanes (POPs) as the most abundant constituents of Marsdenia tenacissima (M. tenacissima) were novel ABCB1-modulatory pro-drugs, which underwent intestinal microbiota-mediated biotransformation in vivo to generate active metabolites. V2O5 nanowires with an intrinsic peroxidase-like activity

V2O5 nanowires exhibit an intrinsic catalytic activity towards classical peroxidase substrates such as 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 3,3,5,5,-tetramethylbenzdine (TMB) in the presence of H2O2. These V2O5 nanowires show an optimum reactivity at a pH of 4.0 and the catalytic activity is dependent on the concentration. The Michaelis-Menten kinetics of the ABTS oxidation over these nanowires reveals a behavior similar to that of their natural vanadium-dependent haloperoxidase (V-HPO) counterparts. The V2O5 nanowires mediate the oxidation of ABTS in the presence of H2O2 with a turnover frequency (k(cat)) of 2.5 x 10(3) s(-1). The K-M values of the V2O5 nanowire…

Dry eye disease affects 15% of the population over the age of 65. It is characterized mainly by inflammation of the ocular surface (tear film, conjunctiva and cornea) which represents a relevant target for therapeutic strategies. The side effects and long-term intolerance of anti-inflammatory drugs give rise to an increasing interest for dietary components exhibiting anti-inflammatory properties, such as polyunsaturated fatty acids (PUFAs). Recognition of oxovanadium(V) species and its separation from other metal species through selective complexation by some acyclic ligands

Acyclic molecules possessing –OH (phenoxo and alkoxo type) groups and imine or amine moieties have been developed to sense the specific preference for VO3+ species. These molecules also showed a capability to quantitatively separate oxovanadium(V) species from a reaction mixture containing metal species of V, Mo, U, Fe, and Mn ions in solution. A cascade quantitative separation of VO3+ followed by cis–MoO2+2 followed by trans –UO2+2 species is demonstrated from their mixture. Synthesis and structural details of oxo-species of vanadium molybdenum and uranium are also discussed. Factors influencing the complexation of these molecules towards oxo metal species of V, Mo and U are also addressed.

The co-metabolism of citrate plus xylose by Leuconostoc mesenteroides subsp. mesenteroides results in a growth stimulation, an increase in D-lactate and acetate production and repression of ethanol production. This correlated well with the levels of key enzymes involved. A partial repression of alcohol dehydrogenase and a marked stimulation of acetate kinase were observed. High citrate bioconversion yields in diacetyl plus acetoin were obtained at pH 5.2 in batch (11.5%) or in chemostat (up to 17.4%) culture. In contrast, no diacetyl or acetoin was detected in citrate plus glucose fermentation.
Inhibitory effects on mitochondrial complex I of semisynthetic mono-Tetrahydrofuran acetogenin derivatives

Modifications in the terminal alpha,beta-unsaturated gamma-methyl-gamma-lactone moiety or in the alkyl chain that links this terminal gamma-lactone with the alpha,alpha'-dihydroxylated THF system of the natural mono-tetrahydrofuranic acetogenins, annonacin and annonacinone, led to the preparation of eight semisynthetic derivatives. Their inhibitory effects on mitochondrial complex I is discussed and compared with that of the classical complex I inhibitor, rotenone.
Chemical initiation for butyl and lauryl acrylate monolithic columns for CEC.

Butyl acrylate (BA)- and lauryl acrylate (LA)-based monolithic stationary phases for CEC were synthesized, using a redox system as initiator of polymerization. BA monoliths were initiated with ammonium peroxodisulfate, whereas LA columns were obtained with lauroyl peroxide as initiator. In both cases, TEMED was used to activate the process. The influence of porogenic solvent composition on both morphological and electrochromatographic properties of the resulting monoliths was investigated. Excellent efficiencies (minimum plate heights of 4.2-6.3 microm for BA columns and 2.6-5.3 microm for LA stationary phases, for a PAHs mixture) were achieved.
