Patterns of K-ras mutation in colorectal carcinomas from Iran and Italy (a Gruppo Oncologico dell'Italia Meridionale study): influence of microsatell…

2006

Background: K-ras mutations are a key step in colorectal cancer progression. Such mutations have been widely studied in case series from Western countries but there are few data on the rate and spectrum of mutations in tumors from countries where the epidemiological features of the disease are different. Patients and methods: Tumor samples from 182 Iranian colorectal cancer patients (170 sporadic cases and 12 HNPCC cases) were screened for K-ras mutations at codons 12, 13 and 61 by sequencing analysis. The cases were also characterized for microsatellite instability at mononucleotide repeats by PCR and fragment analysis, and classified according to microsatellite instability status. The fre…

Niemann-Pick Type C-2 Disease: Identification by Analysis of Plasma Cholestane-3β,5α,6β-Triol and Further Insight into the Clinical Phenotype.

2014

Niemann-Pick type C disease is a rare disorder caused by impaired intracellular lipid transport due to mutations in either the NPC1 or the NPC2 gene. Ninety-five % of NPC patients show mutations in the NPC1 gene. A much smaller number of patients suffer from NPC2 disease and present respiratory failure as one of the most frequent symptoms. Several plasma oxysterols are highly elevated in NPC1 and can be used as a biomarker in the diagnosis of NPC1.Plasma cholestane-3β,5α,6β-triol was evaluated as biomarker for NPC2 by GC/MS and LC-MS/MS analysis. The diagnosis was confirmed by Sanger sequencing and filipin staining. We report three NPC2 patients with typical respiratory problems and a detail…

A novel pro-apoptotic role of zinc octacarboxyphthalocyanine in melanoma me45 cancer cell's photodynamic therapy (PDT)

2018

Abstract Zn-based phthalocyanine acts as drug or photosensitizer in photodynamic therapy (PDT) for the treatment of cancer cells. The activated zinc octacarboxyphthalocyanine (ZnPcOC) reacts with oxygen, to generate reactive oxygen species for the damage of melanoma cancer cells, Me45. This in vitro study aimed at investigating the cytotoxic effects of different concentrations of ZnPcOC activated with a diode laser (λ = 685 nm) on Me45, and normal human fibroblast cells, NHDF. To perform this study 104 cells/ml were seeded in 96-well plates and allowed to attach overnight, after which cells were treated with different concentrations of ZnPcOC (10, 20 and 30 μM). After 4 h, cells were irradi…

Control of Bactrocera oleae by low environmental impact methods: NPC methodology to evaluate the efficacy of lure-and-kill method and copper hydroxid…

2011

The NonParametric Combination (NPC) methodology is proposed to verify the influence of the lure-and-kill method, using Ecotraps (Vioryl SA), on the distribution of Bactrocera oleae (Rossi) (Diptera Tephritidae) infestation in olive groves, and the integration of this method with copper hydroxide sprayings. This approach does not require the verification of any rigid assumption as many parametric methods do. Results of field trials indicate a strict relationship among the efficiency of the lure-and-kill method on time, dimension of the field, and population density of the olive fly. Moreover, the lure-and-kill method alone seems insufficient to provide an efficient control of the olive fly p…

Identification and molecular characterization of a novel mutation in MSH2 gene in a lynch syndrome family

2017

Background and aim of the work: The Lynch Syndrome (LS) is associated with germline mutations in one of the MisMatch Repair (MMR) genes, including MLH1, MSH2, MSH6, PMS2, MLH3 and MSH3. The molecular characterization of mutations in these MMR genes facilitates the pre-symptomatic diagnosis of subjects at risk to develop a colon cancer or a cancer LS-related. Methods: DHPLC and direct sequencing were performed for the mutation detection analysis. Results: In this study, we identified a novel frame shift mutation, the named is c.170delT in MSH2 gene that determined a premature stop codon and consequently, the formation of a truncated protein (p. Val56Glyfs*7). This is a novel mutation, as it …

Isolation of a novel LPS-induced component of the ML superfamily in Ciona intestinalis

2015

ML superfamily represents a group of proteins playing important roles in lipid metabolism and innate immune response. In this study, we report the identification of the first component of the ML superfamily in the invertebrate Ciona intestinalis by means of a subtractive hybridization strategy. Sequence homology and phylogenetic analysis showed that this protein forms a specific clade with vertebrate components of the Niemann-Pick type C2 protein and, for this reason, it has been named Ci-NPC2. The putative Ci-NPC2 is a 150 amino acids long protein with a short signal peptide, seven cysteine residues, three putative lipid binding site and a three-dimensional model showing a characteristic b…

Cilvēka parvovīrusa B19 infekcijas marķieru un aktivīna B klātbūtne pacientiem ar ME/CF

2021

Iespējams, ME/CFS iedalījums varētu būt ar infekciju saistīta saslimšana. Tādēļ šajā darbā tika pārbaudīta B19 infekcijas marķieru klātbūtne un aktivīna B, potenciālā biomarķiera, koncentrācija ME/CFS pacientiem. Pētījuma ietvaros noteica B19 specifisko IgG/IgM klases antivielu klātbūtni, aktivīna B koncentrāciju, lietojot ELISA testus, un B19 NS genoma sekvences nPCR pilnu asiņu un no šūnām brīvas asins plazmas DNS paraugos ME/CFS pacientiem un praktiski veseliem asins donoriem (PVAD). Procedurāla spēles tēlu (NPC) ģenerēšana

2022

Maternal-fetal cross-talk: elucidating the role of mir-30d in endometrial receptivity and pregnancy outcome

2018

Head–Neck Cancer Delineation

2021

Head–Neck Cancer (HNC) has a relevant impact on the oncology patient population and for this reason, the present review is dedicated to this type of neoplastic disease. In particular, a collection of methods aimed at tumor delineation is presented, because this is a fundamental task to perform efficient radiotherapy. Such a segmentation task is often performed on uni-modal data (usually Positron Emission Tomography (PET)) even though multi-modal images are preferred (PET-Computerized Tomography (CT)/PET-Magnetic Resonance (MR)). Datasets can be private or freely provided by online repositories on the web. The adopted techniques can belong to the well-known image processing/computer-vision a…
