Negative Staining of Thinly Spread Biological Samples

2007

Negative staining is widely applicable to isolated viruses, protein molecules, macro-molecular assemblies and fibrils, subcellular membrane fractions, liposomes and artificial membranes, synthetic DNA arrays, and also to polymer solutions. In this chapter, techniques are provided for the preparation of the necessary support films (continuous carbon and holey/perforated carbon). The range of suitable negative stains is presented, with some emphasis on the benefit of using ammonium molybdate and of negative stain-trehalose combinations. Protocols are provided for the single-droplet negative staining technique (on continuous and holey carbon support films), the negative staining-carbon film te…

Cooperative High-Temperature Spin Crossover Accompanied by a Highly Anisotropic Structural Distortion

2016

Spin transitions are a spectacular example of molecular switching that can provoke extreme electronic and structural reorganizations in coordination compounds. A new 3D cyanoheterometallic framework, [Fe(pz)(Au(CN)2)2], has been synthesized in which a highly cooperative spin crossover has been observed at 367 and 349 K in heating and cooling modes, respectively. Mössbauer spectroscopy revealed a complete transition between the diamagnetic and paramagnetic states of the iron centres. The low-spin-to-high-spin transition induced a drastic structural distortion involving a large one-directional expansion (ca. 10.6%) and contraction (ca. 9.6%) of the lattice. Negative thermal expansion along th…

Discriminative features of type I and type III secreted proteins from Gram-negative bacteria

2006

AbstractThe amino acid composition of sequences and structural attributes (α-helices, β-sheets) of C-and N-terminal fragments (50 amino acids) were compared to annotated (SWISS-PROT/ TrEMBL) type I (20 sequences) and type III (22 sequences) secreted proteins of Gram-negative bacteria. The discriminant analysis together with the stepwise forward and backward selection of variables revealed the frequencies of the residues Arg, Glu, Gly, Ile, Met, Pro, Ser, Tyr, Val as a set of strong (1-P < 0.001) predictor variables to discriminate between the sequences of type I and type III secreted proteins with a cross-validated accuracy of 98.6–100 %. The internal and external validity of discriminant…

Modified Halloysite Nanotubes: Nanoarchitectures for Enhancing the Capture of Oils from Vapor and Liquid Phases

2013

We prepared hybrid halloysite nanotubes (HNT/sodium alkanoates) in which the inner cavity of the nanoclay was selectively modified. Physicochemical studies evidenced the interactions between HNT and sodium alkanoates, ruled out clay exfoliation, quantified the amount of the loaded substance, and showed an increase of the total net negative charge, allowing us to obtain rather stable aqueous nanoclay dispersions. These dispersions were exploited as inorganic micelles to capture hydrocarbon and aromatic oils in the vapor and liquid states and were revealed to be nonfoaming but very efficient in encapsulating oils. Here, we have fabricated biocompatibile and low-cost inorganic micelles that ca…

3D Metamaterials with Negative Thermal Expansion and Negative Effective Compressibility

2018

Materials with negative thermal expansion are desired for controlling thermal stresses, but unusual in nature. With two-component metamaterials it is possible to tune the thermal expansion from positive over zero to negative values, even if both components have positive thermal expansion. Using gray-tone laser lithography we fabricate three-dimensional two-component polymer microlattices, exhibiting zero or negative thermal expansion [1].

The structure of gas-filled n-butyl-2-cyanoacrylate (BCA) polymer particles

1995

Abstract The structure of gas-filled poly-[n-butyl-2-cyanoacrylate] (BCA) particles has been demonstrated by negative staining with uranyl acetate, platinum-carbon shadowing of air-dried material and thin sectioning of the aqueous suspension of BCA particles, embedded in water-soluble melamine resin. The polymer shell of the hollow particles possesses a globular outer surface and a smoother inner surface.

Studien zum Vorgang der Wasserstoffübertragung, 52. Zur Kenntnis der elektroreduktiven Spaltung der N - N-Bindung

1978

— 3) Ube…

Rearrangements of Nitrile-Stabilized Ammonium Ylides

2014

Rearrangements of nitrile-stabilized ammonium ylides are useful reactions for the construction of alkylamines, nitrogen heterocycles, or carbonyl compounds. In spite of their attractive synthetic potential, they are much less frequently used than rearrangements of ammonium ylides either carrying charge-stabilizing carbonyl groups or being devoid of additional stabilization of the negative charge. This review focuses on different classes of rearrangements of nitrile-stabilized ammonium ylides as well as their application in synthetic organic chemistry. 1 Introduction 2 [1,2]-Stevens Rearrangements 3 [2,3]-Sommelet–Hauser Rearrangement 4 [2,3]-Stevens Rearrangements 5 Competitive [1,2]-, [2,3…

Intensified Job Demands and Cognitive Stress Symptoms : The Moderator Role of Individual Characteristics

2020

Intensified job demands (IJDs) originate in the general accelerated pace of society and ever-changing working conditions, which subject workers to increasing workloads and deadlines, constant planning and decision-making about one’s job and career, and the continual learning of new professional knowledge and skills. This study investigated how individual characteristics, namely negative and positive affectivity related to competence demands, and multitasking preference moderate the association between IJDs and cognitive stress symptoms among media workers (n = 833; 69% female, mean age 48 years). The results show that although IJDs were associated with higher cognitive stress symptoms at wo…
