Abstract The inhibitory effect of cavernolide, a novel C 21 terpene lactone isolated from the sponge Fasciospongia cavernosa , on PLA 2 and other enzyme activities involved in the inflammatory process was studied. Cavernolide inhibited human synovial sPLA 2 in a concentration-dependent manner with an IC 50 value of 8.8 μM. Besides, this compound decreased in the nanomolar range the myeloperoxidase degranulation process using different stimuli. Cavernolide also inhibited TNFα, NO and PGE 2 production in intact cell experiments. NO and PGE 2 reduction was the consequence of the inhibition on iNOS and COX-2 expression because it did not affect inducible nitric oxide synthase and cyclooxygenase…
Dynamical Scaling in Polymer Solutions Investigated by the Neutron Spin-Echo Technique

Chain dynamics in polymer solutions was investigated by means of the recently developed neutron spin echo spectroscopy. — By this technique, it was possible for the first time to verify unambiguously the scaling predictions of the Zimm model in the case of single chain behaviour and to observe the cross over to many chain behaviour. The segmental diffusion of single chains exhibits deviations from a simple exponential law, indicating the importance of memory effects. Abstract Neutron scattering and computer simulations are powerful tools for studying structural and dynamical properties of condensed matter systems in general and of polymer melts in particular. When neutron scattering studies and quantitative atomistic molecular dynamics simulations of the same material are combined, synergy between the methods can result in exciting new insights into polymer melts not obtainable from either method separately. Neutron scattering methods can be applied to solid polymers in order to investigate the structure of the bulk material as well as the nature of the molecular motion. A review of some recently performed studies is presented. Diffusional motion in crystals of chain molecules has been studied by means of quasielastic scattering for the case of n-C33H68 as an example for the relationship between structure and molecular motion. It can be shown that below the so-called rotational transition the paraffin crystals exhibit two types of molecular motion: a translational jump (τt = 0.5 10-8s) and a 180°-rotational jump (τr = 2.5 10-8s). Chemical structure and internal diffusion within polymer chains in the melt

The single chain dynamics of polydimethylsiloxane in the melt is studied by means of quasielastic neutron scattering. For this polymer the wave vector range 0.03 A−1≦q≦0.30 A−1 covers the regime of universal modes as well as local diffusive processes. A model is described which incorporates the specific chemical structure of the macromolecule and allows to interpret our data in the full wave vector regime. The only parameter which enters the model, the monomer diffusion constantDm, is found to be (1.2±0.2)·10−5 cm2s−1.

The objective of scattering studies on biological macromolecules is to determine molecular conformations and arrangements, particularly those that may contribute to specific biological functions. QENS from polymeric micelles in supercritical CO[sub 2]

We report QENS measurements from PS-b-PFOA aggregates in supercritical CO2. These consist of dense cores of CO2-insoluble polystyrene surrounded by a ‘corona’ of PFOA surfactant molecules whose CO2-philic groups interface with supercritical CO2. Lineshapes are dominated by localized diffusive modes and segmental dynamics of the anchored, finite-length PFOA chains. For Q∼0.6 A−1, we obtain effective diffusion coefficients of ≈0.8×10−6 cm2/sec. At higher Q, a single component is not sufficient as shown by excess intensity on the flanks. For Q>1.5 A−1, the wings reflect contributions due to a distribution of faster, more localized chain modes.

Telechelic α,ω-macrozwitterions — narrowly distributed polystyrene chains oppositely charged at each end — were synthesized via anionic polymerization using direct functionalization with the initiator and the termination agent. The resulting products were purified (zwitterion content ≥ 0,95) and characterized with gel permeation chromatography, viscometry, static and dynamic light scattering and small-angle neutron scattering. In unpolar solvents as toluene, the polymeric zwitterions form clusters with molecular weights depending on concentration. The structure of aggregates of a new bolaform surfactant [α-ω (4,7,10,13-pentaoxa-16-azacyclooctadecane) hexadecane] in neat aqueous solution has been investigated by small-angle neutron scattering. Preliminary data analysis unequivocally indicates that globular micelles are formed. This finding has been confirmed by the analysis of data obtained upon addition of hydrochloric acid. The observed significant reduction of micelle aggregation number has been attributed to the peculiar complexing ability of the two aza-crown ether units in the surfactant molecule; this is in line with information inferred from the complexation of ions into the non alkylated azacrown ethers.
