Selection of distinct Valpha/beta T-cell receptor families during in vivo and in vitro T-cell maturation.

The experimental conditions influencing the use of Valphabeta TCR families were examined in lymph node (LN) cells from peptide-immunized C57BL/6 and Vbeta8.2 transgenic mice. Expanded proportions of Vbeta5, Vbeta8.2, Vbeta9, Vbeta12 and Vbeta14 positive cells and an association of Vbeta8.2 with Valpha11 was found in freshly harvested 8-day or 34-day immune LN cells. In contrast, peptide-specific T-cell lines generated in vitro from 8-day immune lymph node cells were found to be almost exclusively of the Valpha2/Vbeta12 family. Abstract IL-2 influences both survival and differentiation of CD4+ T effector and regulatory T cells. We studied the effect of i.n. administration of Abs against the α- and the β-chains of the IL-2R in a murine model of allergic asthma. Blockade of the β- but not the α-chain of the IL-2R after allergen challenge led to a significant reduction of airway hyperresponsiveness. Although both treatments led to reduction of lung inflammation, IL-2 signaling, STAT-5 phosphorylation, and Th2-type cytokine production (IL-4 and IL-5) by lung T cells, IL-13 production and CD4+ T cell survival were solely inhibited by the blockade of the IL-2R β-chain. Abstract Immunodominance hierarchies operating in immune responses to viral Ags limit the diversity of the elicited CD8 T cell responses. We evaluated in I-Ab+/A2-HHD-II and HLA-DR1+/A2-DR1 mice the HLA-A*0201–restricted, multispecific CD8 T cell responses to the human CMV tegument phosphoprotein pp65 (pp65) Ag. Vaccination of mice with pp65-encoding DNA elicited high IFN-γ+ CD8 T cell frequencies to the pp65495–503/(e6) epitope and low responses to the pp65320–328/(e3) and pp65522–530/(e8) epitopes. To identify endocytic receptors that allow dendritic cells (DCs) to capture and present antigens on major histocompatibility complex (MHC) class I products in vivo, we evaluated DEC-205, which is abundant on DCs in lymphoid tissues. Ovalbumin (OVA) protein, when chemically coupled to monoclonal alphaDEC-205 antibody, was presented by CD11c+ lymph node DCs, but not by CD11c- cells, to OVA-specific, CD4+ and CD8+ T cells. Bakalaura darba mērķis bija analizēt vai notiek nanodaļiņu pārnese uz audzēja šūnām mezinhimālo cilmes šūnu un MDA-MB-231 šūnu 3D kopkultūru modelī. Abas šūnu kultūras tika kultivēts S10 barotnē. Šūnu kultūru diferenciēšanai tika izvēlēts CD90 marķieris. MCŠ tika iezīmētas ar QD655 un pēc 6h inkubācijas tika veikta plūsmas citometrijas analīze. 3D kopkultūras modelis tika izveidots izmantojot PolyHEMA pārklājumu. QD pārnese uz vēža šūnām tika analizēta ar plūsmas citometriju pēc 48h kultivēšanas 3D kopkultūras modelī. 97% MCŠ 2D kultūrā pēc 6h inkubācijas uzņem QD655…

Development of a 3D CZT detector prototype for Laue Lens telescope

We report on the development of a 3D position sensitive prototype suitable as focal plane detector for Laue lens telescope. The basic sensitive unit is a drift strip detector based on a CZT crystal, (similar to 19x8 mm(2) area, 2.4 mm thick), irradiated transversally to the electric field direction. The anode side is segmented in 64 strips, that divide the crystal in 8 independent sensor (pixel), each composed by one collecting strip and 7 (one in common) adjacent drift strips.
