Modelling retention and peak shape of small polar solutes analysed by nano-HPLC using methacrylate-based monolithic columns.

Abstract The development of methacrylate-based monolithic columns was studied for the separation of pharmaceutical hydrophilic compounds in nano-liquid chromatography. The selected polymerisation mixture consisted of 7.5% hexyl methacrylate, 4.5% methacrylic acid and 18.0% ethylene dimethacrylate (w/w), in a binary porogenic solvent (35:35 w/w 1-propanol/1,4-butanediol). The polymer synthesised with this mixture has a good permeability, not excessive back-pressure, and reasonable retention times for polar and non-polar solutes. Pretreatment and fractionation of lignocellulosic barley straw by mechanocatalysis

Abstract This study focuses on the mechanocatalytical process combining dilute acid pretreatment and mechanical processing driven by ball milling. Milled and hydrolyzed barley straw samples are subject to reducing sugar analysis by DNS assay and capillary electrophoresis. Optimization of the saccharification conditions was carried out with two different sulfuric acid concentrations, 0.5 mol kg −1 and 1.0 mol kg −1 , and compared. A significant yield of total reducing sugar (53.4%) was obtained from barley straw impregnated with sulfuric acid (1.0 mol kg −1 ) after milling for only 20 min. Luminometric sub-nanoliter droplet-to-droplet array (LUMDA) and its application to drug screening by phase I metabolism enzymes.

Here we show the fabrication of the Luminometric Sub-nanoliter Droplet-to-droplet Array (LUMDA chip) by inkjet printing. The chip is easy to be implemented and allows for a multiplexed multi-step biochemical assay in sub-nanoliter liquid spots. This concept is here applied to the integral membrane enzyme CYP3A4, i.e. the most relevant enzymatic target for phase I drug metabolism, and to some structurally-related inhibitors.

Structure and luminescence properties of supramolecular polymers of amphiphilic aromatic thioether–peptide conjugates in water

We present the preparation of luminophore–peptide conjugates that self-assemble into supramolecular polymers in neutral buffer. To this end, we have prepared a small library of six conjugates with varying substitution patterns of the aromatic thioethers, as well as varying amino acid sequences. The latter have allowed us to tune the thermodynamic driving force for self-assembly and probe their photoluminescent properties either in the monomeric or polymeric state, while fully avoiding selective solvent techniques or organic solvent mixtures.
