Multiple reaction monitoring for identification and quantification of oligosaccharides in legumes using a triple quadrupole mass spectrometer.

Raffinose family oligosaccharides are non-digestible compounds considered as dietary prebiotics with health-related properties. Hence, it is important to develop highly specific methods for their determination. An analytical method is developed in this study for oligosaccharide identification and quantification using liquid chromatography-tandem mass spectrometry equipped with a triple quadrupole analyser operating in Multiple Reaction Monitoring mode. Raffinose, stachyose and verbascose are separated in a 10-minute run and the method is validated over a broad concentration range, showing good linearity, accuracy, precision and high sensitivity. Biochemical and structural features of a novel cyclodextrinase from cow rumen metagenome.

A novel enzyme, RA.04, belonging to the alpha-amylase family was obtained after expression of metagenomic DNA from rumen fluid (Ferrer et al.: Environ. Microbiol. 2005, 7, 1996-2010). The purified RA.04 has a tetrameric structure (280 kDa) and exhibited maximum activity (5000 U/mg protein) at 70 degrees C and was active within an unusually broad pH range from 5.5 to 9.0. It maintained 80% activity at pH 5.0 and 9.5 and 75 degrees C. The enzyme hydrolyzed alpha-D-(1,4) bonds 13-fold faster than alpha-D-(1,6) bonds to yield maltose and glucose as the main products, and it exhibited transglycosylation activity. Carbohydrates in plant immunity and plant protection: roles and potential application as foliar sprays.

International audience; Increasing interest is devoted to carbohydrates for their roles in plant immunity. Some of them are elicitors of plant defenses whereas other ones act as signaling molecules in a manner similar to phytohormones. This review first describes the main classes of carbohydrates associated to plant immunity, their role and mode of action. The synthesis of linear carbohydrate modified polysiloxanes has been successfully performed by hydrosilation of functionalized and protected carbohydrate derivates. The functionalization (allyl glycosilation) may be applied to a wide variety of reducing carbohydrates. In this study, mono-, di- and oligosaccharides were converted by a simple two-step reaction (β-acetylation and allyl glycosilation) into active compounds, allowing the polymer analogous addition to random poly[dimethyl-co-hydromethyl]siloxanes (hydrosilation). Oligosaccharide recognition by selectins: Synthesis and biological activity of multivalent sialyl lewis-X ligands

Abstract Trivalent sialyl Lewis-X ligands 6–8 anchored onto flexible templates have been synthesized and evaluated as inhibitors of E-selectin and P-selectin mediated cell adhesion in cell culture assays and in vivo. Biological activities in vitro correlated with spacer length and lead to ligands with 3-fold (E-selectin) and 5-fold (P-selectin) improved receptor binding avidity per single tetrasaccharide moiety.

Noncovalent Saccharide Recognition by Means of a Tetrakis(bile acid)-Porphyrin Conjugate: Selectivity, Cooperation, and Stability

Molecular recognition of Glu, Glc 2 -Glc 6 and Mal 3 by a tetrakis(bile acid)―porphyrin conjugate has been studied by using ESI-FTICR mass spectrometry. The bile acid conjugate was observed to form 1:1 noncovalent complexes with saccharides. The conjugate was found to have size-selectivity towards saccharides with three or more glucose residues. The Glc 3 and Glc 4 also formed kinetically the most stable complexes. The electron capture dissociation (ECD) experiments revealed that in complexation of an oligosaccharide three glucose residues interact with the bile acid conjugate, whereas additional glucose residues are susceptible to fragmentation. Xyloglucan-derived oligosaccharides induce ethylene synthesis in persimmon (Diospyros kaki L.) fruit

In this work the effect of injection of xyloglucan-derived oligosaccharides (XGOs) into whole persimmon (Diospyros kaki L.) fruits on ethylene biosynthesis was investigated. Fruits collected during different ripening stages produced low levels of ethylene without a climacteric-like peak. Pretreatment of these fruits with 10 cm 3 C 2 H4 m -3 for 8 h stimulated little or no endogenous ethylene production.
