Th17-related cytokine expression is increased in the bronchial mucosa of stable COPD patients

2009

Background: There is an increased number of activated Tl lymphocytes in the bronchial mucosa of stable COPD patients but are absent studies on Th17 cells and their effector cytokines. Objectives: To investigate the expression of retinoic orphan receptor(ROR)C2, interleukin(IL)-17A,IL-17F,IL-21,IL-22 and IL-23 in bronchial biopsies from patients with stable COPD of different severity(stage 2 to 4)compared with age-matched control subject(smokers with normal lung function and lifelong non-smokers).Methods: The expression of RORC2,IL-17A,IL-17F,IL-21,IL-22 and IL-23 was measured in the bronchial mucosa using immunohistochemistry and/or real time quantitative polymerase chain reaction. Role of Smads in respiratory disease pathogenesis

2008

Transforming Growth Factor beta (TGFβ) cytokine plays an important role in normal pulmonary morphogenesis and function as well as in the pathogenesis of lung diseases. The principal signaling pathway downstream to activate TGFβ is the Smad pathway. Even though many studies have focused on Smads’ structural features and pathway, less is known about the possible relationship between protein and mRNA expression of Smads and lung diseases. This review will focus on Smads and sum up what is know about their role in some respiratory diseases: COPD,asthma end fibrosis

BPCO E OSTEOPOROSI

2013

COPD and osteoporosis are frequently associated because they have common risk factors, including age, smoking habits and reduced physical activity. COPD is also characterized by systemic inflammation, malnutrition (up to cachexia), hypovitaminosis D, and the use of local and systemic corticosteroids, all of which could cause deterioration in bone quality with a consequent increase in fragility fractures. These fractures may further impair mobility and increase disability and mortality. The vertebral and rib fractures can directly further affect lung function and promote exacerbations. As a result, the assessment of fracture risk in patients with COPD is of vital importance from the earliest…

Use of ICS in COPD: From Blockbuster Medicine to Precision Medicine

2017

Chronic obstructive pulmonary disease (COPD) is a major cause of mortality worldwide, whose burden is expected to increase in the next decades, because of numerous risk factors, including the aging of the population. COPD is both preventable and treatable by an effective management including risk factor reduction, prevention, assessment, and treatment of acute exacerbations and co-morbidities. The available agents approved for COPD treatment are long-acting or ultra-long-acting β2-agonists (LABAs) and long-acting muscarinic antagonists (LAMAs) bronchodilators, as well as inhaled corticosteroids (ICS) in combination with LABAs. ICS use has been restricted only to selected COPD patients by th…

“Smoke, Choline-Acetyl-Transferase, Muscarinic Receptors and fibroblast proliferation in COPD

2009

Acetylcholine (ACh), synthesized by choline acetyltransferase (ChAT), and muscarinic M1, M2, and M3 receptors (MRs) are involved in fibroblast proliferation. We evaluated ChAT, MRs, and extracellular signal-regulated kinase (ERK) 1/2 and nuclear factor (NF) κB activation in lung fibroblasts from patients with chronic obstructive pulmonary disease (COPD), control smokers, and controls. Human fetal lung fibroblasts (HFL-1) stimulated with interleukin (IL)-1β, tumor necrosis factor (TNF)-α, and cigarette smoke extracts (CSEs) were evaluated for ChAT and MR expression. We tested the effects of ACh on fibroblast proliferation and its ability to bind fibroblasts from patients with COPD, control s…

Physician's mistakes in the interpretation of spirometry

2012

Background. The most recent ATS/ERS recommendations on lung function testing include a definition of airflow obstruction based on lower limit of normal (LLN) of FEV1/FVC and suggest to measure total lung capacity (TLC) in suspected cases of “pseudo-restriction” (normal FEV1/FVC ratio because of concomitant reductions in FEV1 and FVC), that can conceal airflow obstruction if the subject does not exhale long enough. Aims. To evaluate the skill of physicians in the interpretation of spirometry. Methods. A questionnaire focusing on the interpretation of five spirograms was administered to 127 physicians (aged 25-67yrs; 39% pulmonologists, 20% geriatrics). Correlates of spirometric misinterpreta…

SNPs associated withHHIPexpression have differential effects on lung function in males and females

2019

AbstractAdult lung function is highly heritable and 279 genetic loci were recently reported as associated with spirometry-based measures of lung function. Though lung development and function differ between males and females throughout life, there has been no genome-wide study to identify genetic variants with differential effects on lung function in males and females. Here, we present the first genome-wide genotype-by-sex interaction study on four lung function traits in 303,612 participants from the UK Biobank. We detected five SNPs showing genome-wide significant (P<5 × 10−8) interactions with sex on lung function, as well as 21 suggestively significant interactions (P<1 × 10−6). T…

The German COPD cohort COSYCONET: Aims, methods and descriptive analysis of the study population at baseline

2016

Abstract Background The German COPD cohort study COSYCONET (" CO PD and SY stemic consequences- CO morbidities NET work") investigates the interaction of lung disease, comorbidities and systemic inflammation. Recruitment took place from 2010 to 2013 in 31 study centers. In addition to the baseline visit, follow-up visits are scheduled at 6, 18, 36 and 54 months after baseline. The study also comprises a biobank, image bank, and includes health economic data. Here we describe the study design of COSYCONET and present baseline data of our COPD cohort. Methods Inclusion criteria were broad in order to cover a wide range of patterns of the disease. In each visit, patients undergo a large panel …
