Measurement of T Cell Activation After 16‐hr In Vitro Stimulation with Concanavalin A

2010

A flow cytometry assay that can be used to directly determine the proportion of activated T lymphocytes in human whole blood samples after stimulation with concanavalin A is presented here. Human whole blood is incubated with fluorescently labeled antibodies (against CD3, CD4, CD8, and CD69), erythrocytes are then lysed, and the samples are analyzed using a flow cytometer. The assay presented is able to differentiate between CD4+ and CD8+ T lymphocytes. Thus, it is possible to quantify both lymphocyte populations in parallel, as well as the respective proportions of activated T lymphocytes, all from one sample. An additional advantage of this assay is that it was developed to assay whole bl…

Risk factors associated with adverse fetal outcomes in pregnancies affected by Coronavirus disease 2019 (COVID-19): a secondary analysis of the WAPM …

2020

Abstract Objectives To evaluate the strength of association between maternal and pregnancy characteristics and the risk of adverse perinatal outcomes in pregnancies with laboratory confirmed COVID-19. Methods Secondary analysis of a multinational, cohort study on all consecutive pregnant women with laboratory-confirmed COVID-19 from February 1, 2020 to April 30, 2020 from 73 centers from 22 different countries. A confirmed case of COVID-19 was defined as a positive result on real-time reverse-transcriptase-polymerase-chain-reaction (RT-PCR) assay of nasal and pharyngeal swab specimens. The primary outcome was a composite adverse fetal outcome, defined as the presence of either abortion (pre…

Perforator mapping in the trunk: a guide for freestyle flap surgery and an introduction to the sustainable anatomical study

2012

Introduction: Freestyle Perforator flaps minimize flap harvest morbidity and can be harvested anywhere in the body if perforators anatomy is well known. A study of trunk perforator vessels has been performed on recycled CT scans avoiding cadaver investigations or unnecessary radiation exposure. Materials and Methods: 200 CT scans, performed for the study of liver or pancreatic disease, were examined. No unnecessary radiation was administered. Previous abdominal surgery was reason of exclusion. Short time guided bone regeneration using beta-tricalcium phosphate with and without fibronectin. An experimental study in rats

2020

Background  The aim of this histomorphometric study was to assess the bone regeneration potential of beta-tricalcium phosphate with fibronectin (β-TCP-Fn) in critical-sized defects (CSDs) in rats calvarial, to know whether Fn improves the new bone formation in a short time scope. Material and Methods  CSDs were created in 30 Sprague Dawley rats, and divided into four groups (2 or 6 weeks of healing) and type of filling (β-TCP-Fn, β-TCP, empty control). Variables studied were augmented area (AA), gained tissue (GT), mineralized/non mineralized bone matrix (MBM/NMT) and bone substitute (BS). Results 60 samples at 2 and six weeks were evaluated. AA was higher for treatment groups comparing to …

2022

Water parameters in the 2 years before spawning of F0 (08.02.2016-06.03.2018) and during larval and juvenile phase of F1: Larval period until 17.05.2018 (48 dph, 900 dd) and 01.06.2018 (63 dph, ~900 dd) for warm and cold life condition respectively, for the juveniles until 28.09.2018 (180 dph, ~4000 dd) and 12.02.2019 (319 dph, ~5100 dd) for warm and cold conditioned fish respectively. Means ± s.e. over all replicate tanks per condition. Temperature (Temp.), pH (free scale), salinity, oxygen and total alkalinity (TA) were measured weekly in F1 and monthly in F0; sea water (SW) measurements were conducted in 2017 and 2018. Water parameters during larval and early juvenile phase of F0: Larval…

Managing Multi-center Flow Cytometry Data for Immune Monitoring.

2014

With the recent results of promising cancer vaccines and immunotherapy 1 – 5 , immune monitoring has become increasingly relevant for measuring treatment-induced effects on T cells, and an essential tool for shedding light on the mechanisms responsible for a successful treatment. Flow cytometry is the canonical multi-parameter assay for the fine characterization of single cells in solution, and is ubiquitously used in pre-clinical tumor immunology and in cancer immunotherapy trials. Current state-of-the-art polychromatic flow cytometry involves multi-step, multi-reagent assays followed by sample acquisition on sophisticated instruments capable of capturing up to 20 parameters per cell at a…

Calibration of misonidazole labeling by simultaneous measurement of oxygen tension and labeling density in multicellular spheroids

1995

To correlate misonidazole concentrations and oxygen pressures (Po2) at identical locations within EMT6/Ro multi-cell spheroids (mean diameters +/- SD: 867 +/- 20 microns), Po2 measurements were performed with oxygen-sensitive microelectrodes during incubation of these spheroids with tritiated misonidazole (10 mg/I; 445 microCi/mg). In each individual spheroid, Po2 profiles were correlated with the corresponding spatial distribution of misonidazole as quantified by conventional autoradiography and grain counting. To compare the oxygenation status of spheroids in the measuring chamber with that of spheroids in spinner culture, misonidazole labeling was performed in both environments following…

Pharmacological targeting of the novel β-catenin chromatin-associated kinase p38α in colorectal cancer stem cell tumorspheres and organoids

2021

AbstractThe prognosis of locally advanced colorectal cancer (CRC) is currently unsatisfactory. This is mainly due to drug resistance, recurrence, and subsequent metastatic dissemination, which are sustained by the cancer stem cell (CSC) population. The main driver of the CSC gene expression program is Wnt signaling, and previous reports indicate that Wnt3a can activate p38 MAPK. Besides, p38 was shown to feed into the canonical Wnt/β-catenin pathway. Here we show that patient-derived locally advanced CRC stem cells (CRC-SCs) are characterized by increased expression of p38α and are “addicted” to its kinase activity. Of note, we found that stage III CRC patients with high p38α levels display…
