Piwi Proteins and piRNAs in Mammalian Oocytes and Early Embryos

SummaryGerm cells of most animals critically depend on piRNAs and Piwi proteins. Surprisingly, piRNAs in mouse oocytes are relatively rare and dispensable. We present compelling evidence for strong Piwi and piRNA expression in oocytes of other mammals. Human fetal oocytes express PIWIL2 and transposon-enriched piRNAs. Oocytes in adult human ovary express PIWIL1 and PIWIL2, whereas those in bovine ovary only express PIWIL1. In human, macaque, and bovine ovaries, we find piRNAs that resemble testis-borne pachytene piRNAs. Unequal cleavage requires a highly organised cytoskeleton. We investigated the localisation of both tubulins and microtubular arrays in Dreissena eggs during and after fertilisation using confocal laser scanning microscopy. Freshly spawned eggs are arrested in metaphase I. A maternal pool of γ-tubulin is found mainly in the centre of the asters of the meiotic spindle. The paternal pool of γ-tubulin, present in the fertilising sperm, could not be traced within the egg, but a microtubule-organising centre forms near the male pronucleus at anaphase II. Effect of genistein added to bull semen after thawing on pronuclear and sperm quality.

The aim of this research was to study the effect of different genistein treatments on bull sperm after thawing on pronuclear formation after in vitro fertilization (IVF) and on different sperm quality variables. Three experiments were performed. In Experiment 1, three treatments (Control, sperm incubation for 1 h at 37 degrees C with or without genistein) and two sperm concentrations during IVF (I or 3 x 10(6) sperm/mL) were evaluated to study the influence of genistein on pronuclear formation (PNF). SummaryThe zona pellucida (ZP) is a glycoprotein matrix surrounding mammalian oocytes. Upon fertilization, ZP hardening prevents sperm from binding to and penetrating the ZP. Here, we report that targeted gene deletion of the liver-derived plasma protein fetuin-B causes premature ZP hardening and, consequently, female infertility. Transplanting fetuin-B-deficient ovaries into wild-type recipients restores fertility, indicating that plasma fetuin-B is necessary and sufficient for fertilization. In vitro fertilization of oocytes from fetuin-B-deficient mice only worked after rendering the ZP penetrable by laser perforation. Oxytocin Regulates Neurosteroid Modulation of GABAAReceptors in Supraoptic Nucleus around Parturition

In this study, we investigate how neurosteroid sensitivity of GABAAreceptors (GABAARs) is regulated. We examined this issue in neurons of the supraoptic nucleus (SON) of the rat and found that, during parturition, the GABAARs become insensitive to the neurosteroid allopregnanolone attributable to a shift in the balance between the activities of endogenous Ser/Thr phosphatase and PKC. In particular, a constitutive endogenous tone of oxytocin within the SON after parturition suppressed neurosteroid sensitivity of GABAARs via activation of PKC. The primary end-point of the present study was to verify the effect of PL on the pregnancy rate (PR) of our oocytedonation programme.
