Vascular endothelial growth factor gene polymorphisms in ovarian cancer

Abstract Background. Polymorphisms within the vascular endothelial growth factor (VEGF) gene, the most important regulator of angiogenesis and vascular permeability, were shown to be independently associated with an impaired prognosis in various malignancies. No data have been reported in ovarian cancer. Methods. In the present multi-center study, we examined three common polymorphisms within the VEGF gene ( VEGF +405G/C, VEGF −460C/T, and VEGF +936C/T) in 553 Caucasian patients with ovarian cancer using pyrosequencing. Trisomy 12 and translocation (7;9) in an ovarian immature teratoma.

We report a case of an immature malignant ovarian teratoma with peritoneal implants diagnosed in an 18-year-old woman. The tumor was brought into remission after surgery (three laparotomies) and adjuvant chemotherapy. A residual peritoneal implant showed a mature epithelial and glial configuration. Histologically, the neuroectodermal component was dominant in the original tumor as well as in the metastases, being confirmed by immunohistochemistry and electron microscopy. A stem cell line has been obtained with cell culture, having a germ cell character and a yolk sac configuration. Glutathione, glutathione S-transferases alpha and pi, and aldehyde dehydrogenase are associated with resistance to carboplatin and/or cyclophosphamide in cell lines. Therefore, we examined whether the expression of these factors in ovarian cancer tissue specimens is associated with resistance of the patients to combination chemotherapy with cyclophosphamide/carboplatin. Estradiol, progesterone, 17-hydroxyprogesterone, androstenedione and CA125 were assayed in 25 women (12 oophorectomized before entering the study) with ovarian carcinoma. The data from patients were ordered according to the presence or absence of the gonads. The patients with ovaries (Group A) showed significantly higher levels of estradiol (p less than 0.01), progesterone (p less than 0.01) and 17-hydroxyprogesterone (p less than 0.01) than controls. This difference was not observed between oophorectomized patients (Group B) and controls.
