Direct investigation of viscosity of an atypical inner membrane of Bacillus spores: A molecular rotor/FLIM study

Abstract We utilize the fluorescent molecular rotor Bodipy-C12 to investigate the viscoelastic properties of hydrophobic layers of bacterial spores Bacillus subtilis. The molecular rotor shows a marked increase in fluorescence lifetime, from 0.3 to 4 ns, upon viscosity increase from 1 to 1500 cP and can be incorporated into the hydrophobic layers within the spores from dormant state through to germination. We use fluorescence lifetime imaging microscopy to visualize the viscosity inside different compartments of the bacterial spore in order to investigate the inner membrane and relate its compaction to the extreme resistance observed during exposure of spores to toxic chemicals. The lungs are considered to have an impaired capacity to contain infection by pathogenic mycobacteria, even in the presence of effective systemic immunity. In an attempt to understand the underlying cellular mechanisms, we characterized the gammadelta T cell population following intranasal infection with Mycobacterium bovis bacillus Calmette-Guerin (BCG). The peak of gammadelta T cell expansion at 7 days postinfection preceded the 30 day peak of alphabeta T cell expansion and bacterial count. Abstract Rotation correlation times of three chemically similar cyanine dyes of different sizes in n -alcohol solutions have been recorded at several temperatures by using polarized picosecond spectroscopy. For all three dyes the linear temperature dependencies of τ or on η/ T were observed to be independent of solvent up to viscosities of about 60 cP. The rotational motion of the dyes proceeds at much slower rates than the excited state isomerization in viscous solutions of the same fluidity. Mucho antes de la aparición de las denominadas 'film commissions' ya puede apreciarse un uso del audiovisual, y en concreto del cine, como instrumento de promoción de los destinos turísticos.
