p21 Ras is small protein with GTPase activity that regulates proliferation, differentiation and apoptosis in all cell types. The three major isoforms of Ras (H-, K- and N-Ras) differing only for the last 24 aminoacids have different post-translational modifications that lead to localization in diverse plasma membrane microdomains and downstream activation of alternative pathways of signal transduction. This might explain, at least in part, the different biological effects of the Ras isoforms in the cells. Heterogeneity within and between primary colorectal carcinomas and matched metastases as revealed by analysis of Ki-ras and p53 mutations

Analysis of the genetic status of Ki-ras and p53 in primary colorectal carcinomas and matched colorectal liver metastasis from 30 patients reveals an overall heterogeneity both within and between the two tumoral tissues. Both genes were found mutated with a similar frequency in both tissues; however, identical mutations in primary tumor and matched metastasis were found less frequently in the case of the Ki-ras than the p53 gene. Only in three cases the same p53 and Ki-ras mutations found in the primary tumor were found also in the metastasis. Un nuovo derivato policiclico glicosilato è stato ottenuto partendo dal derivato policiclico iodurato, utilizzando la reazione di Sonogashira.Il composto glicosilato saggiato presso l’NCI è risultato attivo su tutte le 60 linee cellulari tumorali del panel, risultando più attivo degli altri composti policiclici precedentemente saggiati. Il composto induce l’arresto del ciclo cellulare in fase G2/M nella linea cellulare MDA-MB231, fa diminuire i livelli di ciclina B1e Cdc-2 mentre produce un aumento dell’inibitore p21WAF1, una chinasi ciclino-dipendente.

The anti-viral type I interferon (IFN) response is initiated by the immediate induction of IFN beta, which is mainly controlled by the IFN-regulatory factor-3 (IRF-3). The signaling pathways mediating viral IRF-3 activation are only poorly defined. We show that the Rho GTPase Rac1 is activated upon virus infection and controls IRF-3 phosphorylation and activity. Inhibition of Rac1 leads to reduced IFN beta promoter activity and to enhanced virus production. As a downstream mediator of Rac signaling towards IRF-3, we have identified the kinase p21-activated kinase (PAK1). La deficienza dell'enzima 21 idrossilasi (21OHD) è la causa più comune di sindrome adrenogenitale (SAG). Il gene implicato è il CYP21 (6p21.3). La SAG è un problema clinico-sociale in quanto nel 95% dei casi genera l'iperandrogenismo dei neonati di sesso femminile. Il trattamento più effettuato in corso di virilizzazzione dei genitali femminili esterni è di tipo chirurgico (genitoplastica). Il trattamento prenatale in utero è una alternativa medica con il fine di compensare il difetto enzimatico e prevenire l'eccesso di androgeni in corso di morfogenesi fetale.
