With the very recent reports on successful gene therapies for several diseases, the likelihood for abuse of gene transfer techniques in elite sports is rapidly increasing. It is therefore very important to develop valid detection techniques for transgenic DNA (tDNA) with ultimate sensitivity and specificity. To date, three slightly different procedures have been reported to reliably detect tDNA with sufficiently high sensitivity. Two utilize a real-time PCR-based approach and one uses a primer-internal, intron-spanning PCR approach (spiPCR). Application of whole genome amplification for forensic analysis

Abstract Fundamental to most forensic analyses is the availability of genomic DNA of adequate quality and quantity. To perform a multitude of genetic analyses and assays requires a sufficiently large amount of template. However, DNA yield from forensic samples is frequently limiting the extent of genetic typing. A possible solution to overcome this “bottleneck” of forensic and paleoarcheological DNA analyses could be the amplification of the entire genomic DNA prior to locus specific PCR analysis. Whole Genome Amplification appears to be a promising tool to obtain sufficient DNA amounts from forensic samples of limited quantity.

Hypobetalipoproteinemias (HBL) represent a heterogeneous group of disorders characterized by reduced plasma levels of total cholesterol (TC), low density lipoprotein-cholesterol (LDL-C) and apolipoprotein B (apoB) below the 5th percentile of the distribution in the population. HBL are defined as primary or secondary according to the underlying causes. Primary monogenic HBL are caused by mutations in several known genes (APOB, PCSK9, MTP, SARA2) or mutations in genes not yet identified. Familial hypobetalipoproteinemia (FHBL) is the most frequent monogenic form of HBL with a dominant mode of inheritance. It may be due to loss-of-function mutations in APOB or, less frequently, in PCSK9 genes.…

Hearing loss and individual differences in normal hearing both have a substantial genetic basis. Although many new genes contributing to deafness have been identified, very little is known about genes/variants modulating the normal range of hearing ability. To fill this gap, we performed a two-stage meta-analysis on hearing thresholds (tested at 0.25, 0.5, 1, 2, 4, 8 kHz) and on pure-tone averages (low-, medium-and high-frequency thresholds grouped) in several isolated populations from Italy and Central Asia (total N = 2636). Nitrogen levels in grape-juices are of major importance in winemaking ensuring adequate yeast growth and fermentation performance. Here we used a comparative transcriptome analysis to uncover wine yeasts responses to nitrogen availability during fermentation. Gene expression was assessed in three genetically and phenotypically divergent commercial wine strains (CEG, VL1 and QA23), under low (67 mg/L) and high nitrogen (670 mg/L) regimes, at three time points during fermentation (12 h, 24 h and 96 h). The transcriptional inhibitor thiolutin blocks mRNA degradation.

Thiolutin is commonly used as a general inhibitor of transcription in yeast. It has been used to calculate mRNA decay rates by stopping the transcription and then determining the relative abundance of individual mRNAs at different times after inhibition. We report here that thiolutin is also an inhibitor of mRNA degradation, and thus its use can lead to miscalculations of mRNA half-lives. The inhibition of mRNA decay seems to affect the mRNA degradation pathway without impeding poly(A) shortening, given that the decay rate of total poly(A) amount is not reduced by thiolutin. The nonbiting midge Chironomus riparius was used to study the remediation potential and secondary effects of activated carbon (AC, ø 63-200 μm) in PCB contaminated sediments. AC amendments efficiently reduced PCB bioavailability determined by Chironomus riparius bioaccumulation tests and passive samplers. PCBs were shown to transfer from larvae to adults. Lower PCB concentrations were observed in adult midges emerging from AC amended compared to unamended sediments. Because of the high numeric complexity of Geometric Algebra, its use in engineering applications relies heavily on tools for ecient implementations. In this article, we introduce a new quality of Geometric Algebra Computing solutions based on a new compiler for Geometric Algebra Parallelism Programs (GAPP). These programs are already optimized in a sense that only the really needed computations are left.
