Objectifs L’utilisation des immunotherapies anti-PD1 dans le traitement des cancers de la peau et du poumon, s’accompagne d’effets secondaires auto-immuns thyroidiens. L’objectif de cette etude etait de determiner leur frequence et d’identifier les facteurs favorisant leur survenue. Methodes Etude retrospective chez 196 patients traites pour un melanome malin ou un cancer bronchique non a petites cellules (CBNPC) par pembrolizumab ou par nivolumab, entre juillet 2014 et fevrier 2018. Quarante-quatre patients ont ete exclus de l’etude (34 pour antecedents de dysthyroidie et 18 par manque de donnees). Chaperonopathies and chaperonotherapy. Hsp60 as therapeutic target in cancer: potential benefits and risks.

In this minireview we focus on Hsp60 as a target for anticancer therapy. We discuss the new concepts of chaperonopathies and chaperonotherapy and present information on Hsp60 localization in the cell membrane of human tumor cells. We describe novel mechanisms for Hsp60 reaching the extracellular environment that involve membrane-associated stages, as well as data on anti-Hsp60 antibodies found in human sera, both in normal subjects and patients affected by autoimmune diseases. Finally, we discuss possible therapeutic applications of anti-Hsp60 antibodies in cancer treatment, evaluating also side effects on non-tumor cells. Immune-checkpoint inhibitors represent the new standard of care in patients with advanced NSCLC who progressed after first-line treatment. Peripheral immune-checkpoint blockade with mAbs to programmed cell death receptor-1 (PD-1) (either nivolumab or pembrolizumab) or PD-Ligand-1 (PD-L1) (atezolizumab, durvalumab, or avelumab) alone or in combination with doublet chemotherapy represents an expanding treatment strategy for metastatic non-small cell lung cancer (mNSCLC) patients. This strategy lays on the capability of these mAbs to rescue tumor-specific cytotoxic T lymphocytes (CTLs) inactivated throughout PD-1 binding to PD-L1/2 in the tumor sites.
