Randomized, Double-Blind Study of Sonidegib (Lde225) in Patients (Pts) with Advanced Basal Cell Carcinoma (Bcc)
2014
ABSTRACT Aim: The BOLT phase 2 study, comparing 2 doses of sonidegib, a hedgehog pathway inhibitor (HhPI), in pts with advanced BCC (aBCC; NCT01327053), met its primary endpoint of objective response rate ≥30% in both arms in analyses of data collected up to 6 mo after randomization of the last pt (June 28, 2013, cutoff; median follow-up [f/u], 13.9 mo; Migden, ASCO 2014). Associations of GLI1 (marker of Hh pathway activation) with clinical outcome (as of June 28, 2013) and updated 12-mo efficacy and safety data (Dec 31, 2013, cutoff; median f/u, 20.0 mo) are presented. Methods: Pts with locally advanced BCC (LaBCC; n = 194) not amenable to curative surgery or radiation or metastatic BCC (m…
408 Phase I, first-in-human trial evaluating BI 1387446 (stimulator of interferon genes [STING] agonist) alone and combined with BI 754091 (anti-prog…
2020
Background Activation of the STING pathway in intratumoral immune cells leads to increased type I interferon production, promoting recruitment and priming of T-cells against tumor antigens, and providing anti-tumor activity.1 Intratumoral administration of STING agonists has resulted in notable therapeutic activity in animal models.1 STING agonists have also shown clinical activity in patients, which was more pronounced when combined with an anti-PD-1 antibody.2,3 BI 1387446 potently and highly selectively activates the STING pathway; BI 754091 is a humanized IgG4 anti-PD-1 monoclonal antibody. Intratumoral administration of BI 1387446 resulted in tumor regression, and enhanced the activity…
Is There Still a Role for Endocrine Therapy Alone in HR+/HER2- Advanced Breast Cancer Patients? Results from the Analysis of Two Data Sets of Patient…
2020
Background: Different studies suggest that fulvestrant 500 mg every 28 days (HD-FUL) could be an active treatment in HR+ advanced breast cancer (ABC) patients even treated with aromatase inhibitors in the adjuvant setting. The aim of this analysis is to describe the outcome of ABC patients treated with HD-FUL as first-line treatment in terms of median duration of treatment and the overall response rate in a real-world setting. Methods: For the purpose of the present analysis, we considered two data sets of HR+ ABC patients collected in Italy between 2012 and 2015 (EVA and GIM-13 AMBRA studies). Res…
From World3 to the Social Assessment of Technology: Remarks on Science, Technology, and Society
1993
From a well-known point of view, science is thought to constitute a world of objective entities (World3) different from the objective world of material things (World 1) and the subjective world of minds or mental states (World2). This is an old trichotomy, of which Popper is the leading contemporary proponent.
A review of DEA models when the internal structure of the decision making units is considered
2005
Classical Data Envelopment Analysis (DEA) models consider each Decision Making Unit (DMU), whose relative efficiency they evaluate, as a "black box", i.e., its internal structure is disregarded. The paper presents a comprehensive framework of the most advanced theoretical findings in DEA when the internal structure of the. DMUs is taken into account, thus giving directions for novel applications of such methodology and introducing it as a powerful toot for complex processes performance evaluation.
24M Progress Report. Period concerning this report: 01.01.2019-31.12.2019.
2019
During the second year of the project the main purpose was to design and develop smart fibre-reinforced plastic (FRP) plate by using the most environmentally stable nanocomposite (NC) configuration. As a result, within the support of collaboration partner Institute for Polymers, Composites and Biomaterials (IPCB, Portici, Italy) during Mobility#4 four basalt FRP plates were produced: two [0]8 plates impregnated with the epoxy or the NC, and two [0/45/90/-45]2 also impregnated with the epoxy or the NC, and two [0/45/90/-45]2. Additionally, a set of the specimens for the epoxy and epoxy filled with 0.1 wt.% of hybrid nanofiller (carbon nanotubes and nanofibers in the ratio 1:1) was developed …
Gardens on Canvas and Paper: Cataloguing Botanical Abundance in Late Medici Tuscany
2016
What is the purpose of portraying a branch from a pear tree in bloom and fruit, of listing hundreds of citrus varieties or of dissecting a huge truffle? When Cosimo III (1642–1723), the penultimate grand duke of the Medici dynasty, commissioned artists and botanists alike to create comprehensive visual and textual catalogues of Tuscan flora, aesthetics and curiosity seemed to go hand in hand with a demonstration of knowledge and prosperity. This article deals with the collection of botanical paintings by Bartolomeo Bimbi (1648–1730), housed in the Casino della Topaia close to the city of Florence, and with the mainly handwritten work of the grand ducal botanist Pier Antonio Micheli (1679–17…
Mapping the onset and progression of atrophy in familial frontotemporal lobar degeneration
2005
Background: Frontotemporal lobar degeneration (FTLD) may be inherited as an autosomal dominant disease. Studying patients "at risk" for developing FTLD can provide insights into the earliest onset and evolution of the disease. Method: We carried out approximately annual clinical, MRI, and neuropsychological assessments on an asymptomatic 51 year old "at risk" family member from a family with FTLD associated with ubiquitin-positive and tau-negative inclusion bodies. We used non-linear (fluid) registration of serial MRI to determine areas undergoing significant regional atrophy at different stages of the disease. Results: Over the first 26 months of the study, the patient remained asymptomati…
Neuroprotection in Parkinson's Disease: a Realistic Goal?
2010
The current issue of CNS Neuroscience & Therapeutics contains an interesting review by Kinecses and Vecsei [1] on the progress in our knowledge related to the pathophysiological mechanisms of Parkinson's disease (PD) and on the development of putative neuroprotective molecules. Since the seminal discovery by Oleh Hornykiewicz that degeneration of DA neurons within the substantia nigra pars compacta (SNc) and the consequential dopamine depletion in the striatum was the cause of neurological symptoms in PD [2], thousands of reviews have been written on the subject, some of them possibly superfluous. Nevertheless, we found this last work enjoyable in terms of readability and in the way the aut…
Progressive facial hemiatrophy with associated osseous lesions
2007
Progressive facial hemiatrophy (PFH) is a rare condition characterized by the slow, progressive appearance of a unilateral facial atrophy that affects the skin, subcutaneous tissue, muscle and bone. We report the case of a 60-year-old female patient whose cutaneous symptoms commenced in 1987 in the form of a purplish erythema on the left side of her face and neck, which subsequently remitted giving rise to an indurated region in the left maxillary region. Since 1995 until the present day, she has developed facial hemiatrophy on the left side accompanied by progressive osseous reabsorption of the upper maxilla and left mandible with atrophy of soft tissue. The association of the onset of PFH…
