baz1b loss-of-function in zebrafish produces phenotypic alterations consistent with the domestication syndrome

2023

BAZ1B is an ubiquitously expressed nuclear protein with roles in chromatin remodelling, DNA replication and repair, and transcription. Reduced baz1b expression disrupts neuronal and neural crest development. Variation in the activity of baz1b has been proposed to underly morphological and behavioural aspects of domestication through disruption of neural crest development. Knockdown of baz1b in Xenopus embryos and Baz1b loss of function (LoF) in mice leads to craniofacial defects consistent with this hypothesis. We generated baz1b LoF zebrafish using CRISPR/Cas9 gene editing to test the hypothesis that baz1b regulates behavioural phenotypes associated with domestication in addition to cranio…

Prevalence and Risk of Anisakid Larvae in Fresh Fish Frequently Consumed in Spain: An Overview

2019

Anisakidosis is a fish-borne zoonosis caused by parasitic nematodes of the family Anisakidae, of which the species belonging to Anisakis simplex complex are the most representative. It is considered an emerging disease in Spain. The objective of this study is to analyse the presence of larvae in fish frequently consumed in Spanish supermarkets, inferring the risk of infection. In total 1,786 specimens of 9 different fish species, from two geographical origins (Atlantic and Mediterranean), acquired fresh and not eviscerated were examined for anisakid nematodes. Analysis showed that 33.7% of the samples were parasitized by Anisakis larvae. The horse mackerel (Trachurus trachurus) presented th…

Ankk1 Loss of Function Disrupts Dopaminergic Pathways in Zebrafish

2022

Ankyrin repeat and kinase domain containing 1 (ANKK1) is a member of the receptor-interacting protein serine/threonine kinase family, known to be involved in cell proliferation, differentiation and activation of transcription factors. Genetic variation within the ANKK1 locus is suggested to play a role in vulnerability to addictions. However, ANKK1 mechanism of action is still poorly understood. It has been suggested that ANKK1 may affect the development and/or functioning of dopaminergic pathways. To test this hypothesis, we generated a CRISPR-Cas9 loss of function ankk1 zebrafish line causing a 27 bp insertion that disrupts the ankk1 sequence introducing an early stop codon. We found that…

Vibriosis in aquaculture. 16th EAFP Conference, Tampere, Finland, 4th September 2013

2014

Aquaculture in brackish and marine water is growing worldwide (FAO, 2014), including new cultured species. Various Vibrio infections occur, and cause significant problems, in fish, crustacean and shellfish (Austin & Austin, 2007. Vibrio anguillarum, V. salmonicida, V. ordalii and V. vulnificus are among the pathogens that lead to the biggest losses in aquaculture all over the world (Toranzo et al. 2005, Sandlund et al., 2010; Sitjà-Bobadilla et al. 2014).

Pyrosequencing survey of intestinal microbiota diversity in cultured sea bass (Dicentrarchus labrax) fed functional diets.

2014

The routine use of chemotherapy to control bacterial diseases in aquatic populations has resulted in the development and spread of antibiotic resistance. The inclusion of immunostimulants in fish diets (functional diets) is one of the main strategies to solve this threat. This study aimed to analyse the intestinal microbiota of cultured European sea bass (Dicentrarchus labrax) fed two functional diets applying pyrosequencing of PCR-amplified 16S rRNA gene. Quality-filtered reads were assigned to family and genus taxonomic levels using the Ribosomal Database Project classifier. The autochthonous intestinal microbiota of sea bass consisted of two dominant bacterial genera: Dysgonomonas (Bacte…

Gut microbiota changes in rainbow trout, Oncorhynchus mykiss (Walbaum), during organic acid feed supplementation and Yersinia ruckeri infection

2013

Gut microbiota (GM) composition of vertebrates are considered to influence health parameters including immunological parameters in the host but knowledge of these issues in fish are sparse. We have performed a 45 day experimental study elucidating effects of dietary supplementation of organic acids (Mera Cid) and immunostimulants on the GM composition of rainbow trout Oncorhynchus mykiss (Walbaum) and investigated its association with blood parameters and disease resistance (against enteric red mouth disease (ERM) caused the bacterium Yersinia ruckeri). A total of 640 fish received either 1) basic fish feed without organic acids or immunostimulants, 2) fish feed with immunostimulants (β6glu…

Fast and accurate methodology for direct mercury determination in hair and nails

2015

AbstractA solvent-free, easy, fast and waste-free methodology was developed for the determination of total mercury levels in hair and nails. Samples were taken from several volunteers and directly analysed, with levels of mercury in the range between 0.5 and 8 ng/mg. The influence of quantity of hair, as well as the addition of small amount of solvent and the necessity of previous treatment were studied. Also, a history of mercury exposure was provided by the distance of the hair from the scalp of each volunteer, the results of which were correlated with fish consumption. Furthermore, a short study of mercury in nails was carried out and correlated to the results from hair mercury levels. A…
