Tiam1 as a Signaling Mediator of Nerve Growth Factor-Dependent Neurite Outgrowth

Nerve Growth Factor (NGF)-induced neuronal differentiation requires the activation of members of the Rho family of small GTPases. However, the molecular mechanisms through which NGF regulates cytoskeletal changes and neurite outgrowth are not totally understood. In this work, we identify the Rac1-specific guanine exchange factor (GEF) Tiam1 as a novel mediator of NGF/TrkA-dependent neurite elongation. In particular, we report that knockdown of Tiam1 causes a significant reduction in Rac1 activity and neurite outgrowth induced by NGF. Rac1 and PAK1 are upstream of IKK-ε and TBK-1 in the viral activation of interferon regulatory factor-3

The anti-viral type I interferon (IFN) response is initiated by the immediate induction of IFN beta, which is mainly controlled by the IFN-regulatory factor-3 (IRF-3). The signaling pathways mediating viral IRF-3 activation are only poorly defined. We show that the Rho GTPase Rac1 is activated upon virus infection and controls IRF-3 phosphorylation and activity. Inhibition of Rac1 leads to reduced IFN beta promoter activity and to enhanced virus production. As a downstream mediator of Rac signaling towards IRF-3, we have identified the kinase p21-activated kinase (PAK1). Rac1 protein signaling is required for DNA damage response stimulated by topoisomerase II poisons.

To investigate the potency of the topoisomerase II (topo II) poisons doxorubicin and etoposide to stimulate the DNA damage response (DDR), S139 phosphorylation of histone H2AX (γH2AX) was analyzed using rat cardiomyoblast cells (H9c2). Etoposide caused a dose-dependent increase in the γH2AX level as shown by Western blotting. By contrast, the doxorubicin response was bell-shaped with high doses failing to increase H2AX phosphorylation. Identical results were obtained by immunohistochemical analysis of γH2AX focus formation, comet assay-based DNA strand break analysis, and measuring the formation of the topo II-DNA cleavable complex. This data was collected during an short-term incubation experiment in March 2017 that investigated the response of a surface plankton community to upwelling. This experiment was carried in the framework of the SFB754-funded KOSMOS mesocosm study that took place in La Punta, Callao, Peru between February-April 2017. A total of six different treatments were used to disentangle chemical and biological characteristics of deep water that influence surface plankton blooms: 2 different deep water sources with different nutrient concentrations; 3 treatments to distinguish the effects of inorganic nutrients, organic nutrients and deep water microbial populations. Discharge management in fresh and brackish water RAS: Combined phosphorus removal by organic flocculants and nitrogen removal in woodchip reactors

The current study combined P and N removal using organic flocculant chemicals and woodchip bioreactors in both freshwater and brackish water (7 ppm) recirculating aquaculture systems (RAS). The use of carbon (C) containing flocculant chemicals in the process was hypothesized to further stimulate C-demanding N removal (denitrification) in bioreactors. The trial of combined P and N removal consisted of four treatments: freshwater and brackish water RAS with and without the addition of supernatant from flocculation process to the woodchip reactor. Assessing the changing parameters of water quality at different points in the river–reservoir system can help prevent river pollution and implement remedial policies. It is also crucial in modeling water resources. Multivariate statistical analysis is useful for the analysis of changes in surface water quality. Biohiilen vaikutus torjunta-aineiden ja ravinteiden huuhtoutumiseen peltomaassa : esimerkkeinä glyfosaatti ja fosfori

Tässä työssä tutkittiin kasvihuonekokein kolmessa eri lämpötilassa (300, 375 ja 475 ºC) tuotetun koivubiohiilen (80 tn/ha) vaikutusta glyfosaatin, aminometyylifosfonihapon (AMPA) ja fosforin (PO4-P) huuhtoutumiseen maaperästä. Lisäksi esiteltiin glyfosaatin ja fosforin keskinäistä vuorovaikutusta maaperässä sekä biohiilen tuotantoon liittyvien muuttujien vaikutusta torjunta-aineiden ja ravinteiden huuhtoutumiseen. Tutkimuksen mukaan maaperän biohiililisäykset vähentävät glyfosaatin huuhtoutumista keskimäärin 71 % ja AMPA:n huuhtoutumista keskimäärin 36 %. Defying Stereotypes with Nanodiamonds: Stable Primary Diamondoid Phosphines

International audience; Direct unequal C-H bond difunctionalization of phosphorylated diamantane was achieved in high yield from the corresponding phosphonates. Reduction of the functionalized phosphonates provides access to novel primary and secondary alkyl/aryl diamantane phosphines. The prepared primary diamantyl phosphines are quite air stable compared to their adamantyl and especially alkyl or aryl analogues. This finding is corroborated by comparing the singly occupied molecular orbital energy levels of the corresponding phosphine radical cations obtained by density functional theory computations.

Harnessing Molecular Fluorophores in the Carbon Dots Matrix: The Case of Safranin O

The origin of fluorescence in carbon dots (C-dots) is still a puzzling phenomenon. The emission is, in most of the cases, due to molecular fluorophores formed in situ during the synthesis. The carbonization during C-dots processing does not allow, however, a fine control of the properties and makes finding the source of the fluorescence a challenging task.
