Historical ethnopharmacology of the herbalists from Krummhübel in the Sudety Mountains (seventeenth to nineteenth century), Silesia

Background Krummhübel (after 1945, Karpacz) in the Sudety Mountains (now SW Poland) was called “the village of pharmacists”. At the end of the seventeenth century, there were 57 households, of which about 40 were inhabited by herbalists. Krummhübel herbalists were the first in the Sudety region who applied medicinal mixtures for the treatment of various diseases (using, among others, plants, oils, minerals and even viper venom) in contrast to previous herbalists who only indicated the use of individual plant species for specific diseases. Betulinic acid (BetA) is a naturally occurring pentacyclic triterpene isolated from the outer bark of white-barked birch trees and many other medicinal plants. Here, we studied betulinic acid's cytotoxic activity against drug-resistant tumor cell lines. P-glycoprotein (MDR1/ABCB1) and BCRP (ABCG2) are known ATP-binding cassette (ABC) drug transporters that mediating MDR. ABCB5 is a close relative to ABCB1, which also mediates MDR. Constitutive activation of the EGF receptor is tightly linked to the development of chemotherapeutic resistance. BetA inhibited P-gp, BCRP, ABCB5 and mutation activated EGFR overexpressing cells with similar efficacy as their drug-sensitive parental counterparts. …

In the last few years, there has been emerging interest in developing treatments against human diseases using natural bioactive content. Here, the powder of the edible mushroom Pleurotus eryngii var. eryngii was mixed with the normal diet of mice bearing C26 colon carcinoma. Interestingly, it was evidenced by a significant increase in the survival rate of C26 tumor-bearing mice accompanied by a significant increase in Hsp90 and Hsp27 protein levels in the tumors. Drug resistance and the severe side effects of chemotherapy necessitate the development of novel anticancer drugs. Natural products are a valuable source for drug development. Scopoletin is a coumarin compound, which can be found in several Artemisia species and other plant genera.
