Southern and fluorescent in situ hybridization detect three RAPD-generated PCR products useful as introgression markers in Petunia

Fluorescent in situ hybridization (FISH) was used to reveal the intrachromosomal organization of 11 RAPD markers localized on the genetic map of Petunia hybrida. The cloned RAPD markers were analyzed by means of Southern hybridization to determine their level of sequence repetition and their specificity in different Petunia species with 2n=14 and 18 chromosomes. The same probes were then used in FISH experiments. Most of the RAPD clones studied showed high sequence repetition and no species specificity. Characterizing the effects of salt stress in Calendula officinalis L

In this study the effects of salt stress on growth and several stress markers were investigated in the ornamental and medicinal plant Calendula officinalis. One month old plants were submitted to increasing concentrations of salt up to 150mM NaCl for a period of 30 days. Salinity affected growth in terms of stem length and fresh weight of the plants, but water content remained unchanged indicating a certain tolerance to low and mild concentrations of salt. Although Na+ and Cl− increased in parallel to applied salt treatments, the levels of K+ and Ca2+ showed no significant change, while Mg2+ levels recorded a two folds increase upon the application of the highest salt concentration. The European spruce bark beetle Ips typographus L. causes significant economic losses in managed coniferous forests in Central and Northern Europe. New infestations either occur in previously undisturbed forest stands (i.e., spot initiation) or depend on proximity to previous years’ infestations (i.e., spot spreading). The oviposition choice of an insect herbivore is based on a complex set of stimuli and responses. In this study, we examined the effect of plant secondary chemistry (the iridoid glycosides aucubin and catalpol) and aspects of size of the plant Plantago lanceolata, on the oviposition behavior of the specialist butterfly Melitaea cinxia. Iridoid glycosides are known to deter feeding or decrease the growth rate of generalist insect herbivores, but can act as oviposition cues and feeding stimulants for specialized herbivores. Response of grapevine (Cabernet Sauvignon cv) to above ground and subsurface drip irrigation under arid conditions

Abstract The response of wine grapes to irrigation systems was investigated in a Cabernet Sauvignon/140 Ru vineyard in sandy loam soil in Sicily during a two-year study. Two different drip irrigation systems were evaluated: one surface drip and two subsurface drip irrigation systems, with the trickle line located at different distances from vine trunks. Soil and Regulated Deficit Irrigation Affect Growth, Yield and Quality of ‘Nero d’Avola’ Grapes in a Semi-Arid Environment

The present work studied the effect of two consecutive years of regulated deficit irrigation (RDI) compared to rain fed management on the vegetative growth, yield, and quality of ‘Nero d’Avola’ grapes. The trial was conducted separately in two soils (vertisol and entisol) located at the top and bottom hillside of the same vineyard. Vertisol was characterized by greater depth, organic matter, exchangeable K20, and total N than entisol. Water status and gas exchange of pistachio trees under different irrigation levels

Deficit irrigation and maturation stage influence quality and flavonoid composition of ‘Valencia’ orange fruit

Background: Effects of continuous deficit irrigation (DI) and partial rootzone drying (PRD) treatments (50% ETc) in comparison with full irrigation (CI, 100% ETc) were investigated during 'Valencia' orange fruit maturation. Ultra-high-performance liquid chromatography/high-resolution mass spectrometry was used to quantify hesperidin, narirutin, tangeritin, nobiletin, didymin and neoeriocitrin in the fruit juice and peel.; Results: No significant effect of irrigation was found on yield, juice soluble solids or acidity.
