International audience; Introduction : L’incidence des réactions d’hypersensibilité immédiate aux sels de platine (cisplatine, carboplatine et oxaliplatine) augmente avec leur large utilisation en cancérologie. IntraPeritoneal Chemotherapy (IPEC) is commonly used to treat colorectal cancer metastases. However there is no standardized protocol.The aim of this work was to model this chemotherapy in vitro and to understand the role of hypotonic conditions in this model and its impact on cell death.We determined that the optimal treatment parameters on HCT116 human colon cancer cells, were an exposure of the cells for 30 minutes to 400μM of oxaliplatin under hypotonic conditions (G2.5%) at 37 °C. Předložená dizertační práce se zabývá studiem nových katalyzátorů na bázi kov-oxid vhodných pro použití v palivových článcích na straně anody. Platinou dopovaný oxid ceru připravený magnetronovým naprašováním ve formě tenkých vrstev na uhlíkových mezivrstvách nesených křemíkovým substrátem byl zkoumán prostřednictvím mikroskopických a spektroskopických metod. Synthesis of mesoporous silica nanoparticles (MSNs) and encapsulation of cisplatin for targeted cancer therapies

The aim of this PhD thesis is to elaborate mesoporous silica nanoparticles (MSNs) able to sustain the release of cisplatin into the intracellular compartments of solid tumors. The parametric study shows that morphological, structural and textural properties of MSNs-MCM-41, synthesized by sol-gel reaction by using TEOS as a silica source and CTAB as a structure-directing agent, depend on pH, stirring speed, temperature and extraction process of CTAB. Etude des mécanismes d'action d'une immunothérapie par un lipide A, seul ou associé à l'oxaliplatine, dans des modèles de cancers coliques

Colorectal cancer is a major public health concern in France. Resistance to standard chemotherapy requires development of novel therapeutic approaches. In the past decades, our team showed the immunotherapeutic properties of lipid A in a model of colon cancer in rats. 95% of rats bearing small carcinomas were cured following treatment by lipid A. The study of mechanisms underlying this immunotherapy allowed us to show that the antitumor effect of lipid A was dependent on cytotoxicity induced by granzyme B produced by intratumoral neutrophils.
