Objective To assess the antiproliferative, proapoptotic, and antiangiogenic effects of the double-stranded RNA mimic polyinosine-polycytidylic acid (pIC) complexed with polyethylenimine [pIC PEI ] in xenografted human leiomyomas. Design Heterologous leiomyoma mouse model. Setting University-affiliated infertility center. Animal(s) Ovariectomized and hormone-replaced nude mice (n = 16) who received human leiomyoma fragment transplantation. Stability of PEI–DNA and DOTAP–DNA complexes: effect of alkaline pH, heparin and serum

Abstract DNA complexes formed with nonviral vectors such as polyethylenimine (PEI) or 1,2-dioleoyl-3-trimethylammonium-propane (DOTAP) are widely used in gene therapy. These complexes prevent the interaction of DNA with the fluorescent probes usually employed to quantify DNA. We thus studied the procedures for DNA quantification from DNA complexes as well as their stability in the presence of DNase or mouse, rat and human sera. Release of the DNA from its complexes was accomplished by increasing the pH of the medium (from 7.3 to 13.4) or by adding heparin. Synthesis of polymer materials for use as cell culture substrates

International audience; Up to today, several techniques have been used to maintain cells in culture for studying many aspects of cell biology and physiology. More often, cell culture is dependent on proper anchorage of cells to the growth surface. Thus, poly-L-lysine, fibronectin or laminin are the most commonly used substrates. In this study, electrosynthesized biocompatible polymer films are proposed as an alternative to these standard substrates. ABSTRACT Polyethylenimines are cationic polymers with potential as delivery vectors in gene therapy and with proven antimicrobial activity. However, the antiviral activity of polyethylenimines has not been addressed in detail thus far. We have studied the inhibitory effects of a linear 25-kDa polyethylenimine on infections with human papillomaviruses and human cytomegaloviruses. Preincubation of cells with polyethylenimine blocked primary attachment of both viruses to cells, resulting in a significant reduction of infection. Hydrophilic polycations form complexes when mixed with plasmids. Following functionalisation with glycidyltrimethylammonium chloride (GTA) alpha,beta-poly(asparthylhydrazide) (PAHy), a water-soluble synthetic macromolecule, becomes polycationic and potentially useful for systemic gene delivery. Initially the biocompatibility of PAHy and PAHy-GTA derivatives with different degrees of positive charge substitution were studied and it was shown that PAHy-GTA was neither haemolytic nor cytotoxicity up to 1 mg/ml. Effect of media composition on long-term in vitro stability of barium alginate and polyacrylic acid multilayer microcapsules.

For a number of applications stability of microcapsules is a critical factor. Since the maintenance of polyelectrolyte complexes depends considerably on the ion composition we tested the physical properties of barium alginate capsules and searched for conditions to improve stability by a multilayer coating with polyethylenimine (PEI) and polyacrylic acid (PAA). Mechanical stability and diameters were determined in barium alginate capsules and compared with multilayer capsules. Multilayer coating resulted in smaller capsules than barium complexing alone. Hyperbranched polyethylenimines as versatile precursors for the preparation of different type of unimolecular micelles

Abstract Hyperbranched polyethylenimine (HPEI) was successfully employed as precursor for the preparation of unimolecular inverted and aqueous micelles. The unimolecular inverted micelles (UIMs) obtained by the amidation of HPEI with 1,1′-carbonyldiimidazole (CDI) activated palmitic acid exhibited high encapsulation efficiency for hydrophilic anionic dyes and the efficiency could be enhanced significantly by decreasing the degree of amidation or quaternizing the residual amines.
