A simple, rapid extraction and assay procedure for the NAD+-dependent sorbitol dehydrogenase (SDH) in peach

1998

Sorbitol is the major photosynthetic product in peach. In sink tissues, sorbitol is converted to fructose via the NAD+-dependent sorbitol dehydrogenase (SDH). A new procedure is described that allows rapid, simple quantification of SDH activity in growing tissues. The procedure uses only 0.01 to 2.5 grams of fresh tissue per sample, such that a single shoot tip, a single root tip, or about 5 g of fruit flesh can be assayed for SDH activity. Storage of samples at 4 or -20 C° over night resulted in significant loss of enzyme activity. Thus, freshly harvested tissues were ground with sand in buffer at 2°C in a mortar and pestle, and the homogenate centrifuged @ 3000 ´ g to remove particulate m…

Possible role of mannitol as an oxygen radical scavenger in olive

2011

Along with mannose and sucrose, olive produces large amounts of mannitol in photosynthesizing leaves. Leaf mannitol shows large variation among olive genotypes and Sicilian cultivars with different leaf mannitol content were used in our trials to see whether mannitol may function as an additional non-enzymatic system to protect cells from oxygen radicals and photosystem damage. In one experiment, entire shoots of 'Passulunara', 'Castriciana', 'Nocellara del Belice', and 'Moresca' olive trees were taken to a greenhouse, and mature leaves were treated with paraquat solution, which generates oxygen radicals in presence of light. After 48 hours leaves were sampled, photographed and percent of n…

NAOs and vdW‐DF for simulating co‐adsorption of water and polyols on metal surfaces

2015

The computational approach employed in this study is based on the combined use of numerical atomic orbitals (NAOs), which are recognized as highly efficient basis sets, and different parameterization of vdW-DF exchange-correlation functionals, namely DRSLL and KBM as implemented in the SIESTA code.

Effect of Different Matrices and Nanofillers on the Rheological Behavior of Polymer-Clay Nanocomposites

2010

In this work, a comprehensive study of the rheologi-cal behavior under shear and isothermal and nonisothermal elongational flow of low density polyethylene (LDPE) and ethylene-vinyl acetate copolymer (EVA) based nanocomposites was reported to evaluate their "filmability", that is, the ability of these material to be processed for film forming applications. The influence of two different kinds of organoclay - namely Cloisite 15A and Cloisite 30B - and their concentration was evaluated. The presence of filler clearly affects the rheological behavior in oscil-latory state of polyolefin-based nanocomposites but the increase of complex viscosity and the shear thinning are not dramatic. A larger …

Novel silicon microparticles to improve sunlight stability of raw polypropylene

2015

Oxidation of polyolefins by ultraviolet/visible irradiation is a significant limitation for their use in several technological applications. The use of high-tech additives such as silicon microparticles becomes a compositing strategy that can improve the performance of these materials at long-term service conditions. Silicon particles were added to non-additivated raw polypropylene (PP) prepared by hot melt extrusion and subjected to accelerated sunlight irradiation tests. The stability of thermal properties, mechanical performance and thermal decomposition behaviour of composites was evaluated by differential scanning calorimetry, dynamic mechanical thermal analysis and thermogravimetry. T…

Diurnal Regulation of Leaf Water Status in High- and Low-Mannitol Olive Cultivars

2014

The role of mannitol and malic acid in the regulation of diurnal leaf water relations was investigated in ‘Biancolilla’ (high-mannitol) and ‘Cerasuola’ (low-mannitol) olive trees. Photosynthetic photon flux density (PPFD), vapor pressure deficit (VPD), stomatal conductance (gs), transpiration rate (T), relative water content (RWC), mannitol and malic acid were measured in ‘Biancolilla’ and ‘Cerasuola’ leaves during a dry and hot day of summer in Sicily. In general, leaves of ‘Biancolilla’ trees exhibited greater mannitol content, higher gs and T, but lower RWC than leaves of ‘Cerasuola’ trees. Differences in gs and T between the two cultivars were evident mainly in mid to late morning. ‘Bia…

From Mannose to Small Amphiphilic Polyol: Perfect Linearity Leads To Spontaneous Aggregation

2016

Terminally unsaturated and diastereochemically pure polyol derived from d-mannose shows spontaneous aggregation behavior in water solution. In order to study and clarify this unforeseen phenomenon, a conformational study based on NMR spectroscopy combined with ab initio structure analysis using the COSMO-solvation model was pursued. The results, together with X-ray diffraction studies, suggest a low energy linear conformation for this particular substrate both in solid states and in solution. For such small-sized acyclic carbohydrate derivatives, the linear conformation appears to be a key prerequisite for the unusual molecular self-assembly reported herein. peerReviewed

Improved synthesis and application of conjugation-amenable polyols from d-mannose

2019

A series of polyhydroxyl sulfides and triazoles was prepared by reacting allyl and propargyl d-mannose derivatives with selected thiols and azides in thiol-ene and Huisgen click reactions. Conformational analysis by NMR spectroscopy proved that the intrinsic rigidity and linear conformation of the mannose derived polyol backbone is retained in the final click products in solution. Single crystal X-ray structure determination of one of the compounds prepared further verified that the linear conformation of the polyol segment is also retained in the solid state. In addition, an improved method for direct Barbier-type propargylation of unprotected d-mannose is reported. The new reaction protoc…

Investigation of Polyol Adsorption on Ru, Pd, and Re Using vdW Density Functionals

2015

Biomass-based feedstocks are often oxygenated compounds characterized by large amounts of hydroxyls. As an example, polyols and sugar alcohols are largely employed as reactants for different chemical catalytic transformations such as oxidation, dehydration, and hydrodeoxigenation, which usually occur in aqueous medium. With the goal of gaining new insights into processes that could be industrially relevant, the adsorption of the OH groups on metal surfaces and in the presence of water has to be correctly reproduced and described, within a chosen theoretical framework. Toward this goal, several tests were performed on the catalytically relevant metal Ru, Pd, and Re, benchmarking their bulk a…
