Genetic diversity in Cytisus aeolicus Guss. (Leguminosae), a rare endemite of the Italian flora

ABSTRACT Cytisus aeolicus Guss, is an endemic plant restricted to the isles of Vulcano, Stromboli and Alicudi in the Aeolian archipelago. All known populations were assayed for genetic variability using enzyme polymorphism. Allozyme variation at 16 loci coding for 10 enzyme systems was examined. The great majority of loci turned out to be monomorphic or fixed heterozygous. The observed genetic depauperation is indicative of historical factors, such as the bottleneck effect associated with migration, and the founder effect in population re-establishment. Chemical and spectroscopic characteristics of the wood of Vitis vinifera cv. Sangiovese affected by esca disease.

Chemical and spectroscopic analyses ((13)C cross-polarization-magic angle spinning NMR and attenuated total reflection Fourier transform infrared spectroscopies) were carried out on the wood of Vitis vinifera cv. Sangiovese with brown-red discoloration and black streaks caused by esca disease. The analyses of the brown-red wood revealed the destruction of hemicelluloses and noncrystalline cellulose as well as modifications in the pectic and ligninic wood fractions. The pectic fraction consisted of carbohydrates associated with polyphenols. In modern plants, 2.7 to 2.8 m3 of solid volume veneer blocks are consumed to produce 1 m3 of plywood. After the hydrothermal treatment and debarking of blocks, waste bark is obtained, which makes up 12.5% of the wood mass, while 16-20% of bark is composed of birch outer bark (BOB). Recalculating, BOB makes up 2.0-3.4% of the veneer log mass. Bark is currently burned in boiler houses that is not rational. BOB contains large amount of valuable extractives (up to 34% from o.d. A novel method for determination of inorganic polyphosphates using the fluorescent dye fura-2.

A method for determining inorganic polyphosphate, which is based on the Mn2+-induced quenching of the fluorescence of the calcium indicator fura-2, is described. The effect of Mn2+ ions on fura-2 fluorescence is gradually abolished in the presence of increasing concentrations of polyphosphate; this allows the quantification both of synthetic polyphosphates and of the naturally occurring polymer isolated from tissues or cells. The described method has some advantages compared to conventional procedures for detection of polyphosphates based on the metachromatic effect on toluidine blue.
