Targeting the chromosomal passenger complex subunit INCENP induces polyploidization, apoptosis and senescence in neuroblastoma

2019

Abstract Chromosomal passenger complex (CPC) has been demonstrated to be a potential target of cancer therapy by inhibiting Aurora B or survivin in different types of cancer including neuroblastoma. However, chemical inhibition of either Aurora B or survivin does not target CPC specifically due to off-target effects or CPC-independent activities of these two components. In a previous chromatin-focused siRNA screen, we found that neuroblastoma cells were particularly vulnerable to loss of INCENP, a gene encoding a key scaffolding component of the CPC. In this study, INCENP was highly expressed by neuroblastoma cells, and its expression decreased following retinoic acid–induced neuroblastoma …

Allopolyploidisation in a geological collision zone: on the origin of the tetraploid Anthemis cupaniana Nyman (Compositae, Anthemideae) in Sicily

2022

AbstractThe genus Anthemis has a circum-Mediterranean distribution and comprises c. 175 annual, biennial, and perennial species with polyploid species and species complexes found in its section A. sect. Hiorthia. In Sicily, the genus is represented by 13 species, one of these being the tetraploid A. cupaniana, which is endemic to the island and is distributed throughout the limestone mountains at elevations between 500 and 1800 m a.s.l. Discordant positions in phylogenetic trees based on two plastid regions (psbA-trnH and trnC-petN) and on one nuclear marker (nrDNA ITS1 + 5.8S + ITS2) reveal that the species is of allopolyploid origin, with the maternal parent from the species group around …

Diagnostic genetic screening for assisted reproductive technologies patients with macrozoospermia

2017

International audience; Macrozoospermia is characterized by a high proportion of abnormal spermatozoa with enlarged heads. So far, it has been associated with mutations only in the Aurora Kinase C gene (AURKC) in some cases. Although many publications have reported failure to conceive in couples with macrozoospermia, a few others have described successful pregnancies, thus raising questions as to whether ICSI and AURKC genetic screening should be recommended in all patients with macrozoospermia. First, we report on two monozygotic twins presenting macrozoospermia for whom the genetic status was explored (Aurora Kinase C sequencing) and whole semen and gradient-selected spermatozoa were anal…

Cytogenetic findings in secondary acute nonlymphocytic leukemia

1992

Abstract We here report the results of cytogenetic studies carried out in eight patients with acute nonlymphocytic leukemia developed after primary neoplasias. In seven of the reported cases, clonal chromosome aberrations were found, some being specific of de novo acute nonlymphocytic leukemia (ANLL). Numerical abnormalities were detected, such as the total monosomy of chromosomes 5, 7, 21, trisomy of chromosomes 8, 11, 15, and duplication of chromosome Y. Structural changes were also observed: a del(12)(p12), a del(16)(q22), the translocations t(3;5)(p21;q35),t(3;7)(p21;q35), and t(12;14)(p12;q32) and other changes involving chromosome 8. The finding of a hypertetraploid karyotype with com…

DNA image cytometry

1992

In 68 patients the DNA content of tumor cells was measured by image cytometry after resection of the rectum because of cancer. In the DNA histogram a differentiation between diploid (n = 19), polyploid (n = 24), hypotriploid (n = 17), and hypertriploid (n = 8) tumors was possible. The best relapse-free survival time was found in patients with diploid tumors. The prognosis worsened from polyploid to hypotriploid and was worse in hypertriploid tumors. Testing for a prognostic advantage of diploid over aneuploid tumors without adjustment for additional factors simply by means of the log-rank statistic gave a (one-sided) P of 0.1013. In a multivariate analysis the degree of differentiation turn…

Cytogenetics, flow cytometry, cytophotometry and morphometry of 22 cases of primary breast carcinoma. A comparative study.

1992

Cytogenetic, flow cytometric, cytophotometric and morphometric analyses were performed on 22 previously untreated, primary solid breast carcinomas. Although the cell nuclei as the primary object of these studies were the same in all the tumors, distinct features were evaluated in each case to determine to what degree the results obtained by these techniques are comparable. From the cytogenetic viewpoint, six tumors had a modal number in the diploid range, seven were in the triploid range, and two in the tetraploid range; seven tumors had no modal number. These data correlate with the flow cytometry and cytophotometry results obtained, with DNA values slightly higher than their respective ch…

Reciprocal hybridization at different times between Senecio flavus and Senecio glaucus gave rise to two polyploid species in north Africa and south‐w…

2006

Summary • The analysis of hybrid plant taxa using molecular methods has considerably extended understanding of possible pathways of hybrid evolution. • Here, we investigated the origin of the tetraploid Senecio mohavensis ssp. breviflorus and the hexaploid Senecio hoggariensis by sequencing of nuclear and chloroplast DNA, and by analysis of the distribution of taxon-specific amplified fragment length polymorphism (AFLP) fragments. • Both taxa originated from hybridization between the diploid Senecio flavus and Senecio glaucus. Whereas S. glaucus was the female parent in the origin of S. mohavensis ssp. breviflorus, S. flavus was the female parent in the origin of S. hoggariensis. • The dist…

The marbled crayfish as a paradigm for saltational speciation by autopolyploidy and parthenogenesis in animals

2015

ABSTRACT The parthenogenetic all-female marbled crayfish is a novel research model and potent invader of freshwater ecosystems. It is a triploid descendant of the sexually reproducing slough crayfish, Procambarus fallax, but its taxonomic status has remained unsettled. By cross-breeding experiments and parentage analysis we show here that marbled crayfish and P. fallax are reproductively separated. Both crayfish copulate readily, suggesting that the reproductive barrier is set at the cytogenetic rather than the behavioural level. Analysis of complete mitochondrial genomes of marbled crayfish from laboratory lineages and wild populations demonstrates genetic identity and indicates a single o…

2005

Mitotic chromosome numbers are reported from 20 vascular plant taxa that are endemic to the Balearic Islands and poorly known cytogenetically. The chromosome numbers of Arenaria grandiflora L. ssp. glabrescens (Willk.) G. Lopez & Nieto Feliner (2n = 44), Dianthus rupicola Viv. ssp. bocchoriana L. Llorens & Gradaille (2n = 30), Solenopsis minuta (L.) C. Presl. ssp. balearica (E. Wimm.) Meikle (2n = 28), Romulea asumptionis Font Quer & Garcias Font (2n = 56), Scutellaria balearica Barcelo (2n = 22) and Galium balearicum Briq. (2n = 22) have been determined for the first time. A new chromosome number was found in two populations of Helictotrichon crassifolium (Font Quer) Holub (2n = c. 98) sug…
